ObjectiveLaryngeal carcinoma has an increasing incidence these years. Researchers have been focused on investigating pathogenesis, early discovery, accurate diagnosis and proper treatment.Recent studies show that the growth and metastasis of malignant tumor depended on vascular growth. The neovascularity not noly provides sufficient nurtrition for the growth of tumor, but also a good place for draining off metabolite, and a favourable means for metastasis. Therefore,tumor angiogenesis plays an important part in the growth,development,invasion,metastasis and recurrence. CD105,Galectin-3, EphA2 and its ligand EphrinA1 proteins are reported to be involved in angiogenesis, and play an important role in biological behavior of malignant tumors. There is little research on their relevance to malignancy progression of laryngeal carcinoma. We discussed CD105,Galectin-3,EphA2 and its ligand EphrinA1's roles in pathogenesis,pathology,and diagnosis,treatment and prognosis,which will provide an experimental evidence for application of anti-angiogenic therapy in laryngeal carcinoma.MethodsParaffin-embedded sections and clinic data from patients with primary squmamous cell carcinoma of larynx diagnosed and treated at Department of Otolaryngology of the First Municipal people's Hospital of Guangzhou from January 2000 to June 2004 were studied.We respectively used CD105 and Galectin-3 monoclonal antibody,EphA2 and EphrinA1 polyclonal antibody to detect their expressions. Microvessel density (MVD) highlighted by CDl05 staining was counted according to a standard protocol. We also analyzed the correlation between the four proteins and the impact on survival.The SPSS package(17.0) was used for statistical analyses.Results1 The mean of MVD value (10.33±2.29) for CDl05 expression in tumor tissue of 76 cases was markedly increased,compared with that (1.20±1.04) in 25 normol tunica mucosa laryngis tissues(p<0.05). The expression of CD105 was related to histological,T stage,clinical stage classification, and metastasis in cervical nodes in laryngeal carcinoma (p<0.05). The recurrence of tumor was markedly related to MVD,and MVD was related to prognosis of laryngeal carcinoma.2 Galectin-3 protein expression had a significantly positive detetction rate of 86.84% (66/76) in laryngeal carcinoma tissues,compared with that of 36% (9/25) in normol tunica mucosa laryngis tissues( p<0.05).The expression of Galectin-3 was related to T stage,clinical stage classification, and metastases in cervical nodes in laryngeal carcinoma( p<0.05). The recurrence of tumor was markedly related to Galectin-3,and Galectin-3 was related to prognosis of laryngeal carcinoma.3 EphA2 and EphrinA1 protein were expressed in cytoplasm or cytomembrane of cancer cells. EphA2 and EphrinA1 protein expression had a significantly positive detetction rate of 78.95% (60/76) and 81.85% (62/76) in laryngeal carcinoma, compared with that of 40% (10/25) and 44% (11/25) in normol tunica mucosa laryngis (p<0.05).The expression of EphA2 was related to tumor localization,histological,T stage,clinical stage classification,and metastases in cervical nodes in laryngeal carcinoma( p<0.05). The expression of EphrinA1 was related to T stage,clinical stage classification,and metastases in cervical nodes in laryngeal carcinoma( p<0.05). The recurrence of tumor was markedly related to EphA2 and EphrinA1,and EphA2 and EphrinA1 was related to prognosis of laryngeal carcinoma. 4 The expressions of CD105,Galectin-3,EphA2 and EphrinA1 proteins markedly have a positive correlation in laryngeal carcinoma.5 Survival analysis showed that the life span of patients with 1aryngeal carcinoma is significantly inverse-correlated with the number of MVD and the expressions of Galectin-3, EphA2 and EphrinA1 proteins( p<0.05). Cox Regression analysis indicated that expressions of CD105, Galectin-3 and EphA2 were related to life span of 1aryngeal carcinoma patients. The staging of tumor,metastases in cervical nodes,TNM stage and tumor recurrence are independent factors for tumor prognosis in laryngeal carcinoma.Conclusions1 CDl05 is significantly associated with neovascularization in laryngeal carcinoma,and a marker of angiogenesis.Angiogenesis plays an important role in the invasion, metastasis and recurrence of tumors.2 The increased expression of Galectin-3 is significantly associated with the occurrence of 1aryngeal carcinoma and plays a role in the neovascularity,invasion, metastasis and prognosis of 1aryngeal carcinoma.3 EphA2 and EphrinA1 proteins are increasingly expressed in 1aryngeal carcinoma. The expression of EphA2 was related to histological,T stage,clinical stage classification,and metastases in cervical nodes in laryngeal carcinoma. The expression of EphrinA1 was correlative with T stage,clinical stage classification, and metastases in cervical nodes in laryngeal carcinoma. There is a positive correlation between expressions of EphA2 and EphrinA1 proteins. EphA2 and EphrinA1 may be the carcinogenic factors in 1aryngeal carcinoma,which may be the markers for malignancy of 1aryngeal carcinoma.4 The expressions of CD105,Galectin-3,EphA2 and EphrinA1 proteins markedly have a positive correlation in laryngeal carcinoma,which synergistically accelerate the occurrence and growth of 1aryngeal carcinoma.5 Survival analysis showed that the short life span of patients with 1aryngeal carcinoma is significantly related to the increased expressions of Galectin-3,EphA2 and EphrinA1 proteins,which suggested that Galectin-3,EphA2 and EphrinA1 proteins will provide great potentials in prognosis of laryngeal carcinoma.6 The detection of expression of CDl05-MVD,Galectin-3,EphA2 and EphrinA1 may help with the diagnosis of 1aryngeal carcinoma. They could be important indicators for invasion,metastasis and prognosis in laryngeal carcinoma,which could provide experimental evidence for antiangiogenesis by using gene therapy in 1aryngeal carcinoma.
