Entada phaseoloides (Linn. )Merr. belongs to Entada of Leguminosae plants, which widely distributed in tropical America and Africa, about 30 species exist all around the word. There is only one specie in China. Entada phaseoloides is one of the folk medicines in Dai region, Yunnan Province, which has been widely used for the treatment of stomachache, haemorrhoids, hydroncus and constipation. Modern pharmacologic researches found that Entada genus plants have biological activity on anti-bacterial action, anti-oxidant effect, hypoglycemic effect, anti-tumor, anti-inflammatory and so on. In order to develop and use Entada phaseoloides better, the change of chemical compounds, toxicology and pharmacology of the crude and processed products of Entada phaseoloides were studied respectively in this paper.HPLC-UV was proposed for studying the fingerprints of fresh Entada phaseoloides and their processed products, respectively. HPLC-ESI-MS was introduced to analyze the common peaks in each batch crude Entada phaseoloides. Sixteen characteristic peaks were found in crude Entada phaseoloides samples and twenty-one common peaks existed in processed Entada phaseoloides samples, nine characteristic peaks of which were identified by comparison of the retention time and their molecular weights of chemical standards, most of them were identificated belong to triterpenoid saponins and glucosides. After processed, the chemical composition of the content extracted with solution of 60% methanol from crude Entada phaseoloides are less or more than that from processed Entada phaseoloides, and the changes of the main peaks of fingerprint chromatographic suggest that HPLC can be used to reflect the difference of chemical composition of Entada phaseoloides and their processed products, it would be an efficient way for qualitative control of Entada phaseoloides. To determine the content of Phaseoloideside D and Phaseoloideside E from the crude and processed products of Entada phaseoloides with HPLC. The results found the content of Phaseoloideside D and Phaseoloideside E slightly reduced after processing drugs. This method was simple and accurate which can be used for quality control of the crude and processed products of Entada phaseoloides.The extraction and purification of the total-saponins from Entada phaseoloides with HP-20 resin column were studied. Using Rheediinosede B as the control, the total-saponins absorbency was determined by high performance microplate spectrophotometer. The results showed that the method was easy to operate and may help to establish the quality standards for the total-saponins from Entada phaseoloides.Using the method of intragastric administration, to observe the acute toxicity of the crude and processed products of Entada phaseoloides in mice and determine their LD50. With the methods of charcoal propulsion of small intestine and methyl orange colorimetry of gastric emptying, to study the impact of the crude and processed products of Entada phaseoloides on gastrointestinal movement in mice. Antitumor activities were tested in mice with experimental tumor(S180) in vivo. The mice were treated with total-saponins from Entada phaseoloides for 10 d, after that, inhibition ratio, spleen index, thymus index and hepar index were caculated. The lever of superoxide dismutase (SOD) and malondialdehyde (MDA) were detected with high performance microplate spectrophotometer. Mice ear edema caused by xylene and paw edema induced by carrageenin were employed to study the anti-inflammatory effects of total-saponins from Entada phaseoloides High performance microplate spectrophotometer method was used to measure SOD and MDA lever in blood serum in inflammatory mice caused by carrageenin. The oral LD50 of crude Entada phaseoloides, No.1 and No.2 processed products of Entada phaseoloides in mice were 27.17 g/kg, 35.13 g/kg, 42.18 g/kg body weight. Crude and processed products of Entada phaseoloides can significantly promote the enteric propulsion of normal mice, and can significantly counteract the depressing status induced by atropine, but have no influence on the overactive status induced by neostigmine. The high, middle and low-dose of groups showed significant inhibition of the gastric emptying in normal mice. Processed Entada phaseoloides showed effects on the enteric propulsion of normal and depressing mice, can restrain the gastric emptying under normal mice, but its safety is better than crude Entada phaseoloides. The Entada phaseoloides saponins showed a significant anti-cancer activity on S180 implanted in mice, increased the level of SOD and reduced the content of MDA in blood serum in tumor-bearing mice. The anti-inflammatory experiments displayed that Entada phaseoloides saponins could significantly suppress ear swell caused by xylene in mice, and could restrict paw edema induced by carrageenin in mice, and could increase SOD level and reduce MDA level in blood serum in inflammatory tissue, the anti-inflammatory effect may be correlated with increased SOD level and reduced MDA level.
