ObjectiveIn this experiment the remedial effect of Aescinate was investigated in the diffuse brain injury (DBI) with secondary brain insult (SBI) in rats.Methods105 SD rats were randomly divided into three groups: Normal group,control groups,treatment groups. In treatment groups, aescinate was injected into abdominal cavity in doses of 2.5mg/kg after cerebral injury in 0 hour, 24 hour and 48 hour. The control groups were injected with an equal volume of saline in the same time. The treatment groups and control groups were injected for everyday and raise 10 days. Ten days later, the brain reperfusion fixed and frozen section for immunohistochemistry and Nissl dye. IL-1B,TNF-A,ET(endothelin)and NO levels in brain tissues of the 3 groups were measured.ResultAs compared with the Normal group,levels of IL-1B,TNF-A,ET and NO in rat brain tissues in model group significantly increased(P<0.05).In aescinate treatment group compared with the model group,levels of IL-1B,TNF-A,ET and NO in rat brain tissues significantly decreased(P<0.05). After DBI and SBI,the activity of NOS became higher. However, aescinate reversed the changes described above.ConclusionThe neuroprotective effect of B-sodium aescinate might be achieved by inhibiting in flammatory factor,scavenging oxygen free radicals and stabilizing vascular endothelialcells,etc.Aescinate may play a remedial role by influence the level of NO in brain in early stage.
