PartⅠThe Primary Study on Screening of associated Protein in formation of trauma-induced deep vein thrombosis by Proteomics TechnologiesObject Preliminarily investigate traumatic rat serum using proteomics technology,Establish screening method of protein related to early deep vein thrombosis,Seek the protein in relation to early formation of deep vein thrombosis,Successfully find meaningful biomarkers which may assist in early predicting deep vein thrombosis. Methods Build up the animal model of SD-rats with lower limb trauma-induced DVT ,Prepare the serum of uncontroll and control goup for the following-up experiment,Extract the protein in serum,Afterwards,Conduct Two-dimensional electrophoresis , The first electrophoresis is Isoelectric focusing according to PH Value,The second is Vertical Sodium dodecyl sulfate– polyacrylamide gel electrophoresis(2-DE),Apply software to scan the gel figure and seek matched protein spot, Cut the gel spot which you consider meaningful,Dissolve them with trypsin.Acquired peptide is detected and analysed with Mass spectrometer(MALDI-TOF), Peptide samples were analysed with MALDI-TOF/TOF-MS/MS Mass spectrometer again ,Search SWISS-PROT protein database with fingerprint and Peptide sequence information.Generally analyse the formation received ,and we can identify the diverse protein name. Results Seven kinds of protein are successfully determined between uncontrolled group and controlled,six proteins are increased in the uncontrolled group serum, one protein are reduced in the uncontrolled Conclusion:We successfully detect and identify some protein related to deep vein thrombosis ,it hopefully assists us in early diagnosis and treatment of DVT,Complement family are detected and it shows complement also play the role in DVT formation,but it cannot exclude the possibility of inflammation factors activating complement. PartⅡInfluence on thrombosis formation by inhibitiong of complement activation in low limb-traumatied ratsObject To investigate the influence on thrombosis formation of plasma by exhausting complement in rats with traumatic low limb Methods Forty male SD rats were randomly divided into two groups: control groups and cobra venom factor(CVF) treatment groups , Build up the animal model with Femur fracture induced by trauma in both groups, Separately inject both groups of rats with 50ug/kg and 20ug/kg at 12 hours preoperation and 48 hours postoperation, Blood are distilled and collected from Orbital venous at each time to determine the activity of CH50,At 120 hour, Measure the coagulation indicators of the plasma drowed from heart.Rusult: There is no great changes in CH50 Value of the control group and maintained above 90% at all different time,At 1 hour postoperation CH50 Value Of CVF group are rapidly decreased to 9.8% ,it was be no great change and wave at below of 10%,AS for as coagulation is concerned , there is significant change with PT value and APTT value between two groups (p<0.05),NO statistically significance existed with TT value and FIB value between control group and CVF group.(p>0.05) Conclution Complement inhibitor CVF can obviously deplete complement in traumatic rat ,and achieve the purpose of activing complement ,Reduce complement activation on blood coagulation indicators of trauma,Significantly improved the State of post-traumatic coagulation.
