| Objective: To study the efficacy, mechanism and safety of Showshow on simple obesity, and provide scientific evidences for clinical application.Methods: Normal Control was established. Obesity model of mice was established and then divided into four experimental groups: Positive Control was administered sibutramine solution by gavage, Drug Group was administered Showshow extract, meanwhile, Model Control was administered water. The Lee index, weight growth Rate, fat index and blood lipid level of the different groups were measured and analyzed; in anti-fatigue test, swimming time and vitality were recorded; expression level of fatty acid synthase (FAS) in liver of mice was detected by RT-PCR; Safety of Showshow was evaluated by acute attack test.Results:1. Model establishment: The body weight of mice feed with high fat diet was 20% greater than that of mice feed with basic diet, which indicated that model was established successfully.2. Weight-losing function on fat mice model: After administration, body weight of mice in Drug Group was 7% less than that of mice in Model Control, there was a significant difference between Drug Group and Model Control (P<0.01),however, no obvious difference was observed between Drug Group and Positive Control (P>0.05).weight growth rate of Drug Group was 23.71±1.78%,lowest in all experimental groups, had significant differences compared with those of the three other Controls (P<0.05).Lee index of Drug Group was 3.355±0.083,which was significantly lower than that of Model Control (P<0.05),however, no obvious difference was observed between Drug Group and Positive Control (P>0.05).fat weight of Drug Group was 1.16±0.38 g, 47% lower than that of Model Control, had significant differences compared with those of the three other Controls (P<0.05);fat index of Drug Group was 3.50±1.90%,which was significantly lower than those of Model Control and Positive Control (P<0.05).Fatty deposits on the abdomen of Drug Group were less than Controls. Result of HE staining displayed that the size of Mesenteric adipocytes was obviously smaller than those of Controls.3. Spleen-strengthening function on fat mice model: After administration, swimming time of mice in Drug Group was 15.02±2.49 min, which was one time longer than that of Model Control (P<0.05),and 2.6 times longer than Positive Control (P<0.01).4. Lipid-lowering function on fat mice model: After administration, concentration of TG,TCH and HDL-C of mice serum in Drug Control were 0.7378±0.1293 mmol/L,3.2584±0.5060 mmol/L and 2.5447±0.4425 mmol/L, 28.8%,13.3%,14.7% lower than those of Model Control (P<0.05).5. Lipid-lowering function on normal mice: There was no difference (P>0.05) of basic blood lipid level between the two groups. After treatment, concentration of TG and TCH of mice serum in Drug Control were 1.014±0.408 mmol/L and 3.173±0.225 mmol/L, 40% and 29% lower than those of Control (P<0.05). And concentration of HDL-C was 2.014±0.278 mmol/L, 29% higher than Control (P<0.05).6. Expression level of FAS: Result of RT-PCR showed that expression level of FAS mRNA was higher than Model Control, but lower than Normal Control.7. Safety:Acute peros toxicity test showed no toxicosis and death, and maximal tolerance dose was 180 g·kg-1·d-1, converted to crude drug dose of 868.8 g·kg-1·d-1. Tissue damage was not observed in small intestines of mice in Drug Group by HE staining.Conclusion: Our product Showshow followed the theory of traditional Chinese medicine, and implemented weight-losing and lipid-lowering functions by increasing energy expenditure, especially, it performed well in spleen-strenghtening. Showshow did not cause inappetence, diarrhea and intoxication, which was rational for forming a prescription and in accordance with international weight-losing principle, worthing further study. |
