Cerebrovascular disease was the one of most dangerous diseases in our country, The therapy of cerebrovascular disease has become a major problem in medical science. Ischemic stroke is one of the most common cerebrovascular disease, its causes are cerebrovascular stenosis or embolization, blocked blood flow to the brain,ischemia of brain causing pathological reaction. We adopt the brain artery occlusion model (MCAO) to study the treatment of cerebral ischemia, to explores a new therapy of cerebral ischemia. Meanwhile, reactive oxygen species are an important source of damage, we use multiplePCR technology detection the expression differences of 33 Globins in C.elegans which in oxidation environment. we aim to look for new drugs, provide new treatments for cerebral ischemia.To investigate the neuroprotection of hypervolemic hemodilution on cerebral ischemia-reperfusion of the rat brain. Mature Sprague-Dawley rats were used to prepare the focal ischemia and reperfusion insults by middle cerebral artery occlusion. After then, the physiological saline was injected intravenously to induce hypervolemic hemodilution. The volume of injection included 0.5ml, 1.0ml and 2.0ml respectively. Brain slices were stained with 2, 3, 5-triphenylterzolium chloride (TTC), followed with image analysis to estimate the infarct volume. Our data shows that the infarct volume was reduced as the volume of physiological saline increased. The infarct volume was less than control group significantly when the animal received 2.0ml physiological saline injection. Hypervolemic hemodilution could reduce the infarct size in cerebral ischemia-reperfusion injury and therefore show a neuroprotective effect for the ischemic insults.Neuroglobin(NGB) is a recently discovered oxygen-binding respiratory protein that is distinctive expressed in neurons of the mammalian nervous systems.Ischemia can induce the expressed of neuroglobin ,and the over-expressed of neuroglobin can reduce the cerebral ischemia-reperfusion injury. To investigate the neuroprotection of neuroglobin on cerebral ischemia-reperfusion of the rat brain. Neuroglobin could reduce the infarct size in rat cerebral ischemia-reperfusion injury and therefore show a neuroprotective effect for the ischemic insults.Globin are O2-binding heme proteins present in all kinds of organisms, has the typical"three-over-three"helical sandwich structure. There are 33 putative globins in C.elegans, the function of these globins was unknow. To investigate the relationship between these globins in C.elegans and reactive oxygen free radicals. N2 Wild type C.elegans was used, C.elegans was cultured on nematode growth medium(NGM) which contain different concentrations of paraquat(0.3 mmol/L,0.4 mmol/L,0.5 mmol/L) respectively,after five days , the total RNA was extract from C.elegans,then use multiple-PCR technology amplify 33 globins of C.elegans. Integral light density of each bead was calculated by Gel quantitative analysis software,then get the relative expression quantity of every globin compare with actin gene.we got six globin's genes which expression was increased, they are: Globin-7,Globin -12,Globin -19,Globin -22,Globin -24,Globin -31.These genes expression was increased signification then C.elegans was managed by paraquat(PQ), These genes have an potential ability to inhibit the oxidative damage of PQ.
