| Objective: Through observeing the effects of acupuncture to neurological impairment scores,cerebral infarct area ratio,Ngb,Bcl-2,and Bax in rats hippocampus with cerebral ischemia reperfusion injury after different time point,explore the possible mechanism of acupuncture on cerebral protection.Methods: Reference Zea Longa line to establish the middle cerebral artery occlusion(MCAO)model.120 male SD rats were randomly divided into 4 groups:sham-operated group,model group,control point group and acupoint group.And then each group was divided into 24 h(hour)group and 72 h(hour)group,with 15 rats in each group.Completed the treatment,the neurological impairment score was evaluated.TTC staining was used to detect the ratio of infarct area,Western Blot was used to detect the expression levels of neuroglobin in hippocampus.Bcl-2 and Bax detected by immunohistochemistry.Results:1 Neurological impairment scores: 24 h: Compared with the sham-operated group,the scores of the model group were significantly increased(P<0.01);72 h: Compared with the sham-operated group,the scores of the model group increased significantly(P<0.01);Compared with the model group,the scores of the control group and the acupoint group all decreased,scores of the acupoint group decreased more,the difference had statistical significance(P<0.05).Compared with 24 h groups: There was no significant difference in scores of 72 h model groups,and scores of 72 h control group and 72 h acupoint group were significantly decreased(P<0.01).2 Cerebral infarction area ratio: 24 h: Compared with the sham-operated group,Cerebral infarction area ratio of the rats in other groups were significantly higher(P<0.01);72 h: Compared with the sham-operated group,Cerebral infarction area ratio of the rats in other groups were significantly higher(P<0.01);Compared with the modelgroup,the cerebral infarction area ratio the control point group and the acupoint group decreased obviously(P<0.01).Compared with 24 h groups,72 h control point group of cerebral infarction area ratio decrease,the difference had statistical significance(P<0.05).3 Ngb:(24 h 72h): Compared with the sham-operated group,the expression levels of neuroglobin in model group was declined obviously(P<0.01).Compared with the model group,the expression levels of neuroglobin that in control point group and acupoint group was up-regulated(P<0.01).Compared with 24 h groups,the expression levels of neuroglobin in 72 h acupoint group was declined,the difference had statistical significance(P<0.05).4 Bax Bcl-2:(24 h 72 h): Compared with the sham-operated group,the Bcl-2decreased and the Bax increased in the model group,the difference had statistical significance(P<0.01).Compared with the model group,the Bcl-2 increased,the Bax decreased in the control point group and the acupoint group,the difference had statistical significance(P<0.05 or P<0.01).Conclusion :1 Acupuncture can reduce the neurological impairment scores in rats with cerebral ischemia-reperfusion injury,and reduce cerebral infarction area ratio.2 Its protective mechanism for protect the rats with cerebral ischemia reperfusion injury,maybe by up-regulating the expression level of Ngb,inhibit cell apoptosis.3 The protective effect of acupuncture on brain tissue may be related to the time of acupuncture intervention.In a certain time range,the comprehensive efficacy and duration of treatment are positively correlated. |
