Poria cocos is a traditional Chinese medicine which has a long history in China. It is proved that stomach and kidney can benefit from taking Poria cocos. Polysaccharide is one effective ingredient of Poria cocos. In this research, we investigated the effect of polysaccharides of Poria cocos on inflammation and mouse immunity.The expression of IL-6 and TNF-αwere assayed in inflamed RAW264.7 cells treated with polysaccharides of Poria cocos. And we chose KunMing mice, as experimental material, to test the effect of polysaccharides of Poria cocos on mouse immunity.1) We investigated the physical and chemical properties of PS1 and PS2 which were exstracted from Poria cocos. The molecular weight valus of PS1 and PS2 are 9.5×104 and 20.0×104, respectively. After hydrolysis, the major components are galactose,mannose,glucose and sylose which ratio in mole PS1 is 4.7:2.5: 1.6:1.1 while in PS2 is 1.6:6.5:1.3:0.6.2) In this research, the expression of IL-6 and TNF-αwere obviously inhibited by both PS1 and PS2. We set up a model of inflamed cells by stimulating RAW264.7 cell with LPS and then measured the amount of TNF-α,IL-6 protein and mRNA of TNF-αafter treating the inflamed cells with PS1 and PS2, respectively. The results showed that both PS1 and PS2 can sharply decrease the amount of IL-6 and significantly inhibite the TNF-αboth at mRNA level and protein level. So we infer that polysaccharides of Poria cocos are a potential durg for anti-inflammation.3) We also tested the effect of polysaccharides of Poria cocos in vivo. The KunMing mice were given PS1 and PS2 for several days and then assayed their immune function. The results demonstrated that both PS1 and PS2 have the ability to improve the immune function.In conclusion, both PS1 and PS2 have an attractive prospect in research and application both in inhibiting inflammation and improving immune function.
