Objective:1.The object of study is the key industrial production technology of the integrate primary processing with processing of poria cocos,the bulk troditional medicine in anhui province.We focus on the problems of poria cocos quality unevenness caused by dispersed processing in origin and in order to ensure the quality of poria cocos stable and controllable and improve the quality of the decoction pieces.2.The purpose of the research is to select the technology parameters of poria cocos industrialization production to solve the low completeness of slices caused by mechanical cutting system.To establish operation specification of industrialization production and Standard Operation Procedure(SOP)of the integrate of primary processing with processing.3.Establish and perfect the quality control system of poria cocos for providing the help for the processing technology of the integrate of primary processing with processing and provide the basis for evaluating quality of poria cocos synthetically.4.The optimum processing technology and clinical application of poria cocos butyl were studied by the pharmacological study of the preferred poria cocos butyl processing technology and the traditional processing technology.Methods:1.UV spectrophotometry was used,with glucose,poria cocos acid as standards,to determine the concentration of poria cocos polysaccharide and total triterpenes in poria cocos.HPLC was used to determine the content of grifola acid C,poria cocos to hydrogen acid and poria cocos acid.2.By the quality analysis of poria cocos decoction pieces bought in market and tracing investigation of poria cocos,study different processing methods such as respectively remove the peel while fresh,after sweating,after steamed,sweating steamed,cutting,drying processing link,to find the main factors to influence the quality of poria cocos decoction pieces.3.Poria cocos polysaccharide,total triterpene,grifola acid C,poria cocos to hydrogen acid,poria cocos acid are considered as the index of quality evaluation.Combining with the appearance of the pieces,forming rate and using orthogonal design method to select the best processing mode,to determine the key technical parameters of poria cocos.In addition,the pilot production and fingerprint chromatography were used to establish poria cocos standard operation procedures(SOP).4.Effects of different processing technique poria cocos on diuretic effects in Rats by water load model and metabolic cage.Study on the effect of different processing techniques of poria cocos butyl on spleen deficiency syndrome in rats by dahuang decoction,dropping method and hunger method.Results:1.Research established the industrial mode of the integrate of poria cocos primary processing and processing.The industrial processing technology of poria cocos slice are:the first excavation of fresh poria cocos,sweating until the surface become brown,less surface left over after peeling red skin(that is sweating for 3 days),than cutting into 4-millimetre thick slice,after placing in the shade for two days,put into 50? fan dryer until to be dry.2.Combined with the appearance and molding rate of the poria cocos decoction pieces,the index of poria cocos polysaccharide,total triterpene,grifola acid C,poria cocos to hydrogen acid,and poria cocos acid are used to control the quality of poria cocos decoction pieces synthetically.The weight coefficient of appearance,active ingredients and forming rate of weight coefficient respectively are 0.3,0.5,0.2.The weight coefficient of poria cocos polysaccharide,total triterpene,poria cocos acid,hydrogen to poria cocos acid and grifola acid C respectively are 0.1,0.1,0.1,0.1.In addition,membership degree =(index value-the minimum value of index)/(maximum index-index minimum),comprehensive score = the sum of product of membership degree and weight coefficient,accurate and stable,the method can effectively control the quality of poria cocos.3.The article established the fingerprints chromatography of cporia cocos decoction pieces bought in market and its processing technology.The similarity of bought poria cocos decoction pieces is between 31.1% ~ 99.2% by the fingerprint similarity analysis.The result further illustrate the quality of sold poria cocos decoction pieces is uneven.The similarity of poria cocos process slices is between 98.9% ~ 99.7%,and the similarity of poria cocos cube process slices is between 96.5% ~ 98.8%.Fingerprint technology verified the good reproducibility and high stability of poria cocos decoction processing technology.4.There is a certain pharmacodynamic differences on the diuretic and spleen in different processing technology poria cocos butyl.Poria cocos butyl new technology group diuretic effect is stronger than the traditional group of poria cocos butyl.And poria cocos butyl new technology group than the traditional group of drug absorption effect is good,but no significant difference.Conclusion:1.The article has established quality control method and the comprehensive evaluation method of poria cocos,combining with appearance and decoction pieces molding rate of poria cocos to comprehensively evaluate decoction pieces quality for providing the basis for integrated technology optimization of poria cocos.2.Compared with the traditional process by Manual cutting,the integrate of primary processing with the processing of poria cocos by using machines instead,time-saving and manpower-saving.3.The optimized industrial introduction technology of poria cocos slice and cube is scientific and reasonable,not only improve the forming rate of slices,slices of the process of standardization,industrialization,but also to ensure the quality's stability and controllability,so that the industrialization of poria cocos decoction pieces production can come true.4.Fingerprint chromatography method established is stable and can be used to control the quality of poria cocos slice and poria cocos cube.5.The establishment of water load model rats diuretic test and rat spleen deficiency model spleen test method is scientific,reasonable and feasible,and provides the basis for the clinical application of poria cocos pieces.
