Objective:To investigate the effect of nitric oxide on human umbilical vein endothelial cell apoptosis cultured on different glucose concentration and the relationship between NO and diabetic vascular disease.Methods:Human umbilical vein endothelial cells (human umbilical vein endothelial cells, HUVECs) were cultivated in different concentrations of glucose for 48 hours, and then affected by nitric oxide donor --- sodium nitroprusside (Sodium Nitroprusside, SNP), continue for 24 hours, Flow cytometry(FCM) were used to detect the apoptosis of HUVECs, RT-PCR was used to analysis the expression of Thioredoxin-interacting Protein (Txnip) mRNA.Results:1.Annexin V / PI double staining flow cytometry shows the cell apoptosis rate of high glucose group was significantly higher than the 11.1mmol/l group (P <0.05). NO intervention obviously decreased the cell apoptosis rate caused by high glucose (P <0.05).2.RT-PCR analysis shows that thioredoxin interacting protein (Thioredoxin-interacting Protein, Txnip) mRNA expression was weaker in the NO intervention groups compared with their respective group that doesn't receive NO (P <0.05).Conclusion:As the glucose concentration in the culture medium increased, HUVECs apoptosis rate increased. The mechanism maybe account for increased oxidative stress caused by high glucose which then actives a series of signal transduction pathways to promote the process of cell apoptosis. No can block the oxidative stress and inhibite the endothelial apoptosis induced by high glucose.The mechanism may be related to the decrease of Txnip mRNA expression.
