Reversing Effect Of Curcumin On Multi-drug Resistance Of Human Hepatocellular Carcinoma Bel7402/5-Fu Cell Line

Objective:To investigate the reversing effect of curcumin on Bel7402/5-Fu cell line.Methods: 1.Through exposure to gradual increased concentration of Fluorouracil(5-Fu),the cell line Bel7402 was induced to form a multidrug resistant Sub-cell line Bel7402/5-Fu;2.The sensitivity to chemotherapeutics of Bel7402 and Bel7402/5-Fu were tested by MTT assay, calculating 50% inhibitory concentration(IC50) and resistant index(RI); Differences of inhibition ratio of Bel7402/5-Fu arranged in four groups(the normal control group, the curcumin treated group, the 5-Fu treated group, the curcumin with 5-Fu treated group ) by MTT assay;3.Changes to apoptotic cell content in Bel7402/5-Fu cell in four groups were analyzed by the flow cytometry.Results: The Bel7402/5-Fu cell line showed cross-resistance to Fluorouracil (5-Fu),Adriamycin(ADM),Methotrexate(MTX),cyclophosphamide(CTX),Mitomycin (MMC), and Cisplatin(CDDP),among which 5-Fu had a highest RI(109.55±14.30). Inhibition ratio of Bel7402/5-Fu treated with combination use of 5-Fu and different concentration of curcumin was(21.47±1.49)%,(27.10±2.32)%,(59.37±2.45)% respectively. Apoptosis of Bel7402/5-Fu treated with combination use of 5-Fu and different concentration of curcumin was(30.92±2.10)%,(44.87±2.24)%,(50.36±2.58)%,which was better than curcumin or 5-Fu.Conclusion: Curcumin could induce the apoptosis obviously and have a effective Reversing effect on multidrug resistance of the Bel7402/5-Fu cell line.
