ObjectiveTo study the effects of imidazolines(SP-4-84) combined with cisplatin(DDP) on chemotherapeutic sensitization of hunman liver carcinoma cell lines HepG2 in vitro.Methods1.Human liver carcinoma cell lines HepG2 cells were cultured in vitro;2.The hunman liver carcinoma cell lines were treated with DDP and SP-4-84 singly and both of them,estimating the inhibition of proliferation by MTT-test;3.Cell apoptosis was determined by flow cytometry;4.Detecting the expressions of apoptosis associated gene proteins:Survivin and Bcl-2 by immunohistochemical technique.Results1.The inhibition rate of proliferation of HepG;2.Cells was increased evidently by cisplatin(DDP) combined with SP-4-84(P<0.05). The higher concentration of cisplatin and SP-4-84(6μl) induced the highest rate of apoptosis,the difference was significant compared with group of single cisplatin and the group of cisplatin and the lower concentration of SP-4-84(3μl) (P<0.01);3.By qualitative and quantitative analysis of immunofluorescence images,the expressions of Survivin and Bcl-2 were down-regulated by SP-4-84.Conclusions1.SP-4-84 can enhance chemotherapeutic sensitization for hunman liver carcinoma cell lines HepG2 when combined with cisplatin;2.SP-4-84 inhibit the expressions of apoptosis-associated gene proteins:Survivin and Bcl-2.
