ObjectiveDetecting the phosphatidylinositol 3 - kinase (PI3K) and its downstream molecule protein kinase B (Akt) expression in preeclampsia placenta and Using fasting plasma glucose (FPG) and fasting insulin (FIN) to insulin resistance index (HOMA-IR), To investigate correlation between PI3K and Akt and resistance in the pathogenesis of preeclampsia for the early prevention of preeclampsia to provide a theoretical basis forearly treatment.MethodsChose 30 preeclampsia inpatients( 15 of severe preeclampsia and 15 of mild preeclampsia) of late pregnancy of Jinan municipal women and children's hospital,and 30 normal late pregnancy women as the control. Study conditions: Select without liver, kidney disease, non-essential hypertension, hereditary diseases. In the current pregnancy 30 patients were diagnosed preeclampsia as the case group, Diagnostic criteria: "Obstetrics and Gynecology" Seventh Edition. All the cases were excluded from medical complications and other placenta previa, for example placental abruption, premature rupture of membranes and other obstetric complications. The expression of PI3K and Akt in the placenta of pregnant women were analyzed by immunohistochemistry and positive cells were counted under a microscope. Fasting insulin (FIN) Was measured by radioimmunoassay.Fasting plasma glucose(FPG)Was measured by oxidase assay. Insulin resistance index was calculated using homeostasis model assessment(HOMA)to analyze the relationship between these markers and insulin resistance. (1) The age and height of the preeclampsia group and the normal prognancy group were not statistically (P >0.05). The weight of the preeclampsia group and the normal prognancy group were statistically(P< 0.05) .(2)The levels of FINS and FPG and insulin resistance index were calculated according to homeostasis model assessment in preeclampsia group were significantly higher than those in normal prognancy group(P <0.01) .(3) The level of PI3K protein expression in preeclampsia Group was lower than that in normal pregancy group (P< 0.01).(4) The Akt in preeclampsia group was lower than that in normal pregancy group (P< 0.01).(5) Preeclampsia group PI3k and Akt expression and HOMA-IR were negatively correlated(P<0.01) .(6) The weight of newborns from pregnant women are lighter than the weight of newborns from normal mothers. (P < 0.05).Conclusion(1) Changes in protein expression of PI3k and Akt in endometrial may be one of the molecular mechanisms leading to insulin resistance in patiant with preeclampsia.(2)The levels of FINS and FPG and insulin resistance index were calculated according to homeostasis model assessment in preeclampsia group were significantly higher than those in normal prognancy group.(3) Pregnant women with preeclampsia and abnormal expression of PI3K and AKt contribute to insulin receptor signals to the obstacle, induced an important molecular mechanism of insulin resistance.(4)The weight of newborn of mothers with preeclampsia is less than that of the normal mothers,suggesting that preeclampsia prone to low birth weight children.(5)Increasing protein expression of PI3k and Akt of insulin resistance to lower levels be helpful for the prevention and treatment of the hypertensive disorder complicating pregnancy.
