Expressions Different And Clinic Significance Of GP73 In The Tissue And Serum Of Hapetocellular Carcinoma Patients

Objective: The experimental study choose hepaticcellular carcinoma(HCC),cholangiocellular carcinom(aCCC) , hepatic hemangioma,colonic carcinoma, breast cancer, spuamouscell carcinoma of lung and 40 normal controls as our object, using immunohistochemistry (IHC) and double antibody enzyme linked immunosorbent assay(Elisa), to investigate the expression of GP73 in hepatocellular carcinoma (HCC) tissues and its serum, compare the different expression of GP73 in the tissues and its serum ,to explore the clinic significance of diagnosis HCC .Methods : The expression of GP73 in 80 cases of HCC,10 cases of cholangiocellular carcinoma ,20 cases of hepatic hemangioma,20 cases of colonic carcinoma,20 cases of breast cancer, 20 cases of spuamouscell carcinoma of lung and 20 cases of normal controls were assessed by immunohistochemistry(IHC), the normal controls tissue belongs to hepatic hemangioma ; All of the samples and 4 cases of secondary carcinoma of liver,30 cases of hepatitis B, 20 cases of hepatic cirrhosis,4 cases of hepatitis C and 40 cases of normal controls of the serum were determined by double antibody enzyme linked immunosorbent(Elisa), by using statistics analysis, to detect the different expression of GP73 in the tissues and serum, and detect the relation of expression of GP73 in the serum and clinicopathological parameters, to explore the clinic significance to diagnosis the HCC;Results: 1,GP73 were expressioned in the HCC,cholangiocellular carcinoma,hepatic hemangioma,colonic carcinoma,breast cancer,spuamouscell carcinoma of lung and normal controls. The positive ratio are 85%(68/80),40%(4/10),15%(3/20),25%(5/20),15%(3/20),35%(7/20),15%(6/40) respectively. 2,GP73 were significantly higher in patients with HCC compared to other diseases and normal controls (P<0.05 respectively); 3,Using the normal controls and other diseases as the reference value, the upper limit is 89.1μg/l, the sensitivity and specificity of diagnosis HCC are 73% and 56% of GP73 by it ; 4,The correlation between tissues and serum present positively related(r= 0.705,P<0.05), 5,GP73 expression was significantly related to hepatic cirrhosis, ascites, clinicopathology staging and vessel invasion(P<0.05 respectively). It was irrelevant among expression of GP73 and ages,genders and tumor number (P>0.05).Conclusions: 1, The expressions of GP73 are high in HCC tissues and serum , especially higher in HCC; 2, Using the normal controls as the reference value, the upper limit is 74.4μg/l , and it the upper limit is 89.1μg/l by using the normal controls and other diseases, the sensitivity and specificity of diagnosis HCC are 73% and 56% ; 3, The expression of GP73 was significantly related to the hepatic cirrhosis, ascites, clinicopathology and vessel invasion. 4, GP73 is one of a serum tumor markers of HCC, could be used as for diagnosis of HCC.
