Objective:To investigate the diagnostic value of ovarian cancer by the combination of osteopontion (OPN) and lysophosphatidic acid (LPA).Method:twenty-eight patients suffered with ovarian cancer were collected and served as observed group (OG). Meanwhile,36 patients with benign ovarian tumor in the same period served as control group (CG). The diagnosis of the patients in two groups and the clinic stage of ovarian cancer in observed group were confirmed by post operative histopathologic examination and the manifestation of ovarian cancer. Plasma level of LPA, and serum level of OPN and CA125 were detected by using biochemistry method, ELISA (enzyme linked immunosorbent assay) and CLIA (chemiluminescent immunoassay) respectively. The results of LPA, OPN and CA125 were analyzed with SPSS 17.0 statistical software.Results:1. The plasma level of LPA was 5.25±1.63μmol/L and serum level of OPN was 214.03±117.78 ng/ml in observed group, while the results of LPA and OPN in control group were 1.74±0.82μmol/L and 23.94±5.80 ng/ml respectively. Significant difference exist between two groups for LPA and OPN (both P<0.05). The sensitivity and specificity for detection of ovarian cancer using mixtures of LPA and OPN were 92.86%(26/28) and 83.33%(30/36) respectively, and both of the sensitivity and specificity were significantly better than that of CA125 (both P<0.05). Furthermore, the sensitivity of the combination of OPN and LPA also was significantly better than the single use of LPA or OPN respectively (all P<0.05). However, no significant difference was found for specificity when compared with the combination or single use of LPA or OPN (both P>0.05).2. The plasma level of LPA was 4.12±1.36μmol/L and the serum level of OPN was 149.46±40.89 ng/ml in the early-stage of ovarian cancer patients. However, the results of LPA and OPN were 6.23±1.17μmol/L and 389.03±70.26 ng/ml in late-stage. The sensitivity for early detection of ovarian cancer by the combination of LPA and OPN was 82.5%, while 57%for CA125, significant difference exist between them (P<0.05).3. The area of LPA and OPN under the ROC curve in ovarian cancer group was 0.869 and 0.845 respectively.Conclusion:These results demonstrate that LPA combined with OPN can detect ovarian cancer with high sensitivity, especially for ovarian cancer in early-stage, and encourage further studies to explore clinical values of LPA and OPN.
