Data show that accumulation of cholesterol in the liver cells can cause celldamage,To investigate the injury and the possible mechanisms of cholesterol loadon the hepatic cell,in vitro we tested activation of the unfolded protein response,inflammation and other changes in liver cells under the conditions of cholesterolloading,as well as the correlation between them.L02 cells were incubated 24 h,48 h in 0,50μg/mL,100μg/mL cholesterolalcohol solution respectively;analysis the main marker of UPR (BiP,CHOP),inflammatory factors (CRP,IL-6,TNF-α) mRNA expression levels use RT-PCR;3mM UPR inhibitor PBA were added to different cell groups,and observe everyIndicators expression changes.The results showed that compared with the controlgroup:the cellular UPR marker mRNA expression was positively correlated withcholesterol concentration and Incubation time.UPR also induce the expression ofCRP,IL-6,TNF-α,every factors mRNA expression were detected after adding UPRinhibitor PBA into different groups.Not increase with the corresponding groups ofPBA compared:UPR marker of the mRNA expression was reduced and cut themRNA expression of inflammatory cytokines compared with no PBA groups.Theseresults suggest that L02 cell cholesterol loading can induce endoplasmic reticulumstress, activate the UPR,and the UPR may cause and exacerbate the cells of theinflammation,leading to cell damage eventually....
