Objective: To investigate the effcets of different concentration of urokinase(UK) with stereotactic aspiratopn technique on dissolving rabbit cerebral hematoma volume and heme oxygenase-1 (HO-1) expression around the hematoma after experimental intracerebral hemorrhage model, and discuss the optimal concentration of UK on hematoma dissolve.Method: This experiment used 50 New Zealand rabbits, both male and female,weight 2.5-3.0 kg. Experimental rabbit intracerebral hemorrhage models were made by injected of 0.5ml autologous whole blood. Rabbits were random performed 5 groups: group 1 was control group,injected 0.1ml normal saline(n=10); group 2 to group 5, rabbits were injected different concentration of UK into the hematoma, the concentration of UK included 5000U/ml, 10000U/ml, 30000U/ml, 50000U/ml(n=10/group). Aspiration the dissolution of hematoma 3 hours later. Each group was equally divided into 2 parts, in the first part, the rabbits were investigated the hematoma volume after aspiration, in the seconed group, evaluated the water content of the brains and the expression of the HO-1.Results: Compared with the control group, with the increasing of the UK concentration, the volume solution of the hemorrhage increased gradually, the dissolved hematoma volume of UK groups significantly higher than the control group(P<0.05). The most dissolve effection was 50000U/ml group. Compared with the control group, the brain water content was decreased significantly with the concentration of the UK increasing before 30000U/ml, but increased at 50000U/ml group. 50000U/ml group has a significantly difference compared with 10000U/ml and 30000U/ml groups(P<0.05). With the concentration of UK increasing before 30000U/ml, the expression of HO-1 gradually decreased but increased at 50000U/ml, the lowest expression was the 30000U/ml group.Conclusion: In the experimental rabbit intracerebral hemorrhage models made by injected of 0.5ml autologous whole blood, the concentration of UK was 50000U/ml can significantly increase the dissolution of rabbit brain hemorrhage volume, and the brain water content increased, HO-1 positive expression also increased. The lowest brain water content and the lowest expression of positive cell of the HO-1 was the 30000U/ml group.
