Objective: This experiment is based on the intracerebral hemorrhage models made by injected autologous blood to implement stereotactic hematoma aspiration. The aim of this study is to investigate the effects of different reserving time of filling urokinase on removal of the rabbit intracerebral hemotomas, the brain water content and the heme oxygenase -1 (HO-1) expression. To study the optimized treatment time of hematomas dissolved with urokinase.Methods: 40 healthy New Zealander rabbits, take blood from the rabbits'ear to make intracerebral hemorrhage models. The model rabbits were randomly divided into 4 groups after the operation of 3 hours, implement stereotactic hematoma aspiration: the group A (n=10) was stereotactic operation of hematoma extraction after 1 hour of hematoma cavity injected with urokinase(50000U/ml); the group B (n=10) was stereotactic operation of hematoma extraction after 3 hours of hematoma cavity injected with urokinase; the group C (n=10) was stereotactic operation of hematoma extraction after 6 hours of hematoma cavity injected with urokinase; the group D (n=10) was stereotactic operation of hematoma extraction after 12 hour of hematoma cavity injected with urokinase. 5 rabbits from each group were used to measure the volume of the hematoma, the rest were used to measure the brain water content and heme oxygenase-1(HO-1) expression.Results:1. Volume of hematoma: Implement stereotactic hematoma aspiration after rabbits intercerebral hemorrhage model was established. With the increasing of the reserving time of urokinase, the volume of hematoma become smaller. The volume of hematoma in group A was significantly smaller than that in other groups(P<0.05), while there was no significant difference between other groups.2. Brain water content: With the reserving time of urokinase increased, the brain water content was decreased. The brain water content in group B after urokinase was injected,at the lowest level, then raised slowly. But there were no significant differences among the four groups.3. The expression of HO-1: The count of HO-1 was lower in group B than that in group A, but with the reserving time increased, the expression of HO-1 gradually increased. There was significant difference between group B and group D(P<0.05).Conclusion: 1. The stereotactic hematoma aspiration can decrease the hematoma volume obviously on rabbit cerebral hemorrhage model made by injected of 0.5ml autologous blood. This method had the accurate localization and it was simple. 2. Hematoma dissolved obviously when the reserving time of urokinase(50000U/ml) over 3 hours. 3. The brain water content and the expression of HO-1 were lower in 3 hours group than that in other groups. The results suggest that implement stereotactic hematoma aspiration on intracerebral hemorrhage model, the optimal reserving time of urokinase is 3 hours.
