Objective To investigate the clinical significance of Ki67 expression,DNA ploidy,cell cycle and squamous cell carcinoma of the tongue with different pattern of invasion.Methods 43 cases with histologically proven squamous cell carcinoma of the tongue were classified into four groups accrording to the pattern of invasion described by Anneroth, Ki67 expression was detected respectively by using immunohistochemical staining(SP) and Flow cytometry(FCM) was used to measure DNA ploidy and synthesis phase fraction (SPF).Results Ki67 expression in the tumor cell was varied from the pattern of invasion.The expression in squamous cell carcinoma of the tongue with patternⅢandⅣof invasion had a higher level than that of patternⅠandⅡ(p<0.05). The clinical findings showed that squamous cell carcinoma of the tongue with patternⅢand IVof invasion were easily involved in the cervical lymph node metastasis. DNA ploidy and SPF in the tumor cell were varied from the pattern of invasion, with the pattern of invasion development, the incidence of aneuploid tumors and the SPF gradually increased, the difference statistically significant (p<0.05).Conclusion Pattern of invasion can be considered to be one of effective factors for evaluating the malignance grading of squamous cell carcinoma of the tongue, the higher pattern of invasion and the evidenter expression of Ki67, DNA ploidy and SPF were varied from the pattern of invasion, with the pattern of invasion development, the incidence of aneuploid tumors and the SPF gradually increased. It will be helpful for evaluating the cervical lymph node metastasis and providing the evidence for clinical treatment.
