Chapter 1 The expressions of BDNF/TrkB in Tongue squamous cell carcinomaObjective:To investigate the expressions of brain-derived neurotrophic factor (BDNF) and its primary receptor tyrosine kinase receptor B (TrkB) in 50 cases of Tongue squamous cell carcinoma (TSCC) and 13 cases of normal tissues, analyze the relationship between BDNF/TrkB expression and the clinical pathological features in tongue squamous cell carcinoma.Methods:The expressions of BDNF and TrkB in TSCC tissues obtained from 50 patients and normal tissues from 13 patients were detected by Immunohistochemistry.Results:Immunohistochemical results showed that normal tongue mucosa tissue were all negative for BDNF and TrkB expression,however,in tongue squamous cell carcinoma,the positive rate were 64%(32/50) and 68%(34/50).Positive expression rate of BDNF and TrkB in TSCC were significantly higher than that in normal tongue mucosa (P<0.05). In addition, the rate of positive expression is related to infiltration depth.lymph node metastasis,clinical stage and so on in tongue squamous cell carcinoma.Conclusion:BDNF and TrkB were over appeared expression in tongue squamous cell carcinoma,but were not appeared in normal tongue mucosa,the positive expression of BDNF and TrkB had related with infiltration depth,lymph node metastasis,clinical stage.Chapter 2 Effect of BDNF in proliferation,invasion and metastasis, apoptosis ability of tongue squamous cell carcinomaObjective:To explore the effect of BDNF in proliferation invasion and metastasis, apoptosis ability of tongue squamous cell carcinoma.Methods:The experimental group:control group(Ong/mlBDNF),low concentration group(50ng/mlBDNF),middle concentration group(100ng/mlBDNF),and high concentration group(200ng/mlBDNF);(1)The expressions of BDNF and TrkB in tongue squamous cell carcinoma cell lines (Tca8113 and Cal27) were conducted by immunofluorescence technique;(2) The effect of BDNF on proliferation ability of tongue squamous cell carcinoma cell lines Tca8113 and Cal27 were detected by CCK8 assay;(3)The effect of BDNF on invasion and metastasis ability of Tca8113 and Cal27 were detected by Transwell chamber assay;(4)The effect of BDNF on apoptosis ability of Tca8113 and Cal27 were detected by Flow cytometry instrument Annexin V/PI double staining notation assay.Chi-square test and analysis of variance were used for statistical analysis with SPSS 17.0 software package.Results:BDNF and TrkB are mainly expressed in the cytoplasm of Tca8113 and Cal27.Compared with the control group,200ng/ml of BDNF obviously promoted the proliferation ability of Tca8113 (P<0.05);Compared with the control group,high,medium and low concentration of BDNF can promote the proliferation ability of Cal27,dependence with time and concentration (P<0.05);Compared with the control group,high、medium and low concentration of BDNF can promote the invasion and metastasis ability of Tca8113 and Cal27, dependence with concentration (P<0.05); Compared with the control group,high,medium and low concentration of BDNF can promote inhibit apotosis ability Tca8113 and Cal27,dependence with concentration (P<0.05)Conclusion:BDNF plays an accelerate role on the proliferation,invasion and metastasis,inhibit apoptosis ability of tongue squamous cell carcinoma.
