BackgroundOral squamous cell carcinoma is the most common malignant tumor, surgical treatment is still present is an important part of tongue cancer cure, but surgery on the patient's appearance and function will cause a greater impact, so look for more effective and safe, drug side effects of anticancer drugs has been a cancer drug development workers pursued the goal.Endostatin is an endogenous angiogenesis inhibitor,can effectively prevent proliferation and migration of vascular endothelial cells, but also can promote apoptosis of tumor cells, thus inhibiting tumor growth and metastasis,with strong anti-tumor properties,and is particularlyimportant that the endostatin gene can be specifically targeted and sta-ble endothelial cell lines,treatment of tumors with high efficiency,low toxicity, no drug resistance,etc.,which are the radiation and chemotherapy deficiencies.This study is based on the research status of in vitroexperiments recombinant human endostatin on the tongue squamous cell carcinoma Tca-8113 inhibition for the future and provide experi-mental evidence of its clinical application,and squamous cell carcinom-a of the gene and drug treatment to provide new ideas and theoretic-al basis. ObjectiveRecombinant human endostatin in vitro study of tongue squamous cell carcinoma Tca-8113 cell growth inhibition effect, and clarify its mechanism, to provide experimental basis for clinical application.Materials and methodsCultured tongue squamous cell carcinoma Tca-8113 cells were divided into control and experimental groups; experimental group cells containing different concentrations of endostatin (10umol/L, 20umol/L,30umol/L,40umol/L,50umol/L) Cultured medium, cells cultured in the control group,to terminate the training specified time, the targets weredetected. Cell morphology was observed by inverted microscope, thechange; MTT growth inhibitory effect was detected;flow cytometry analysis cycle and apoptosis;basement membrane reconstruction of experimental observation of endostatin on cell migration; immunocytochemical staining Survivin,MMp-2 gene expression, statistical analysis soft were application Spss13.0, using repeated measures analysis of variance,test for statistical analysis.Results1. With recombinant human endostatin concentration and time increas-ed,its effect on cell growth,significantly enhanced proliferation in cells under inverted microscope shows slow growth and proliferation, cell shrinkage and round shape,off the wall More and more cells, floating in the medium.2. MTT assay showed that endostatin inhibitory effect on cells in adose and time dependent manner.3. With recombinant human endostatin concentration, the cell cycle G1 phase cells gradually increased from 57.67% up to 76.36%; S phase cells increased from 29.07% down to 22.94%. Different conc entrations of endostatin on the role of cell G1 phase to prevent si -gnificant difference.4. With recombinant human endostatin concentration and time increa-sed,the proportion of apoptotic cells gradually increased from 5.32%up to 27.63%. Between different concentrations and different rates of apoptosis between the time the changes are significant differences.5. the role of recombinant human endostatin Tca-8113 cells, cells apoptosis gene Survivin and tumor invasion and metastasis related g-ene expression of MMP-2 decreased compared with control Survivin,MMP-2 in the cytoplasm of the color reduced Coloring in the cytoplasm to reduce that angiostatin can inhibit MingNeipi Tca-8113 cells,Survivin and MMP-2 expression.6. Basement membrane reconstitution experiments showed that the recombinant human endostatin for 48 hours, the concentration of 30umol / L inhibited cell migration of its Tca8113 was 67.94%; concentration of 50mol / L migration inhibitory rate reached 95.12%, and dose-dependent relationship between the concentration.Conclusion1. recombinant human endostatin in vitro can inhibit cell proliferation Tca8113, the inhibitory effect of endostatin concentration, reaction time are dependent.2. Tca8113 recombinant human endostatin can change cell cycle distribution, with a clear role in G1 arrest, and with the concentration, the effect of the more significant.3. Recombinant human endostatin Tca8113 induced apoptosis, and dose -, time - effect relationship. 4. Recombinant human endostatin can down Tca8113 cells, Survivin, MMP-2 expression.5. Recombinant human endostatin inhibits Tca8113 cell migration, thereby inhibiting tumor metastasis, growth.
