Maxillofacial region malignant tumor holds about 10% of all and squamous cell carcinoma is the most common, which is above 80% of it. The therapy level of rumor is developing fast in our country, but it mostly depends on operation, chemotherapy, radiotherapy and immunotherapy etc. The tumor thermotherapy (Hyperthermia) is the fifth treatment method following above-mentioned. With further investigation of biological function to high temperature and improvement of thermotherapy technology, thermotherapy has already played an important role in the complex tumor treatments as the auxiliary treatment method of radiotherapy and chemotherapy. The antitumor effect is remarkable of Taxol (TAX) as a new drug that has been applied into clinic in recent years. It is a kind of peculiar medicine depending on cell cycle, which mainly acts in the G2/M period of the tumor cell. TAX is tubulin-binding agent, which can stabilize microtubules against depolymerization and make the cell proliferation stop in the metaphase. At present, the clinical application of TAX is mainly jointly to treat the tumor with other medicines, but it is less known combined with hyperthermia to treat tumor. For the reason, this research usestongue squamous cell carcinoma cell Line Tca8113 as the research object to investigate the activity of anticarcinoma and the capacity of inducing apoptosis of chemotherapy and thermochemotherapy in vitro with proliferation assay of cells and FCM.We expect to provide the theoretical basis for clinical therapeutic alliance of malignant tumor. Material and methods:1. Research object: Tongue squamous cell carcinoma cell Line Tca8113, cultivated commonly using RPMI-1640 culture medium containing 10% foetus cattle serum.2. According to the different temperature of heating(39℃, 41℃ 和 43℃)and the different concentration of Paclitaxel (1.25, 2.5 , 5, 10 和 20ng/ml) ,the test was divided into 15 groups.Meanwhile five 37℃ control groups were designed at the different concentrations. The apoptosis rate of Tca8113 cell line was analysed with MTT assay.3. Morphological observation: Oserve the growth state and morphology of cells of each group in above-mentioned medicine dosage in the board under inverting microscopes and taking pictures before MTT assay.4. Flow cytometry (FCM) was to measure distribution of the cell cycle by only chemotherapy and heating(41 ℃ )combined with drug.5. Process the data with SPSS 10.0 software package. One way ANOVA was used to analyze means of multiple groups. LSD was for the multiple comparison. Rates were compared with each other by chi-square test. ( a =0.05).Results:1 .Effects on the rate of cell apoptosis(1) Compared with each 37℃ group of the same dosage, the cell apoptosis rate in 41℃ group decreased significantly (P<0.05, P<0.01) .(2) Compared with each 39℃ and 43 ℃ group of the same dosage, the cell apoptosis rate in 41 ℃ group decreased significantly (P<0.05, P<0.01) except the 39℃ and 1.25ng/ml group (P>0.05) .(3) Compared with 37℃ control group at 10ng/ml of TAX, the cell apoptosisrate in 39℃ and 43℃ group decreased significantly (P<0.01) . 2.Effects on cell cycle(1) The distribution of cell cycle changed in only chemotherapy group and heating (41 ℃ )combined with drug group.The distribution of S and G2/M phase increased gradually (P<0.05) .(2) Compared with only chemotherapy group,the distribution of cell cycle has nostatistics significance in heating(41 ℃ )combined with drug group (P>0.05 ) .Conclusions:1 Heating can reinforce the harm of TAX to the Tca8113 cell.At low dosage chemotherapy, 41℃ can raise the inhibition of TAX obviously,which is in coordination with chemotherapy steadly as the optimal tempture.2 Only certain density and special tempture can reinforce the harm of TAX to cell, unsuitable conditions may accelerate the growth and metastasis of tumor cells.The density and tempture should be controlled in the combination of TAX and thermotherapy.
