Objective Observed the influence of three clear hypoglycemic compound onα-glucosidase activity and understood whether the activity ofα-glucosidase is inhibited, we explorated initially its mechanism of hypoglycemic effect, to further provide a theoretical basis for clinical application.Methods1. In vitro experiment: create a vitro filter model ofα-glucosidase activity inhibitory. Different concentrations of water-extraction preparation and ethanol-extraction preparation of three clear hypoglycemic compound were used to drug intervention. With acarbose for the positive drug control group,it was observed on affectα-glucosidase activity.2. In vivo experiment: 70 healthy male WISTAR rats, weighing 180±20g, 10 rats were randomly selected as normal control group (N), and always fed with normal diet. The remaining 60 rats were fed high-fat diet for 4 weeks, with a small dose of streptozotocin (STZ) intraperitoneal injection of 2 consecutive modeling, select the successful model in rats by 50, based on a fasting blood glucose≥11.1mmol/L, the use of randomization method, the rats were divided into 5 groups: model control group (M), Three Clear Hypoglycemic Compound of low dose group (SD), Three Clear Hypoglycemic Compound of middle dose group (SZ), Three Clear Hypoglycemic Compound of high dose group (SG) and acarbose group (A), each group of 10 rats. Normal group and model control group fed with pure water, Three Clear Hypoglycemic Compound high,middle and low group were 8.05,4.03,1.61g/kg rats weight were fed with Three Clear Hypoglycemic Compound, acarbose group fed with acarbose 10.79mg/kg. The day before irrigating stomach, respectively, irrigating stomach of the 2nd week, the first 4 weeks of irrigating stomach, we detected fasting blood glucose, blood glucose after half-hour and two-hour.Results1. In vitro experiments showed that: water-extraction preparation and ethanol-extraction preparation of three clear hypoglycemic compound strongly inhibitedα-glucosidase activity, showed marked concentration - effect relationship. Concentration of 25mg/ml water extract product of three clear hypoglycemic compound onα-glucosidase inhibitory activity of acarbose 2.5mg/ml quite.2. Vivo study showed that(1) 4 weeks after irrigation stomach, blood glucose can be seen between the groups was statistically significant (P<0.05). SD fasting blood glucose level decreased, not including the normal group, pairwise comparison showed, SD group and M group, SD group and A group showed a statistically significant difference (P<0.01), SZ and A statistically significant difference seen in group ( P<0.05).(2) 4 weeks after irrigation stomach, blood glucose after 2hr in each group shows a statistically significant difference (P<0.05). SD group postprandial blood glucose levels were significantly decreased two hours, not including the normal group, pairwise comparison showed, SD group and the SG, M, A group showed a statistically significant difference (P<0.05).(3) 4 weeks after irrigation stomach, the glucose area under the curve between the two groups shows a statistically significant difference (P<0.05). the area under the glucose tolerance curve of SD group significantly reduced, not including the normal group, pairwise comparison showed, SD group and the M, A group showed a statistically significant difference (P<0.05).Conclusion1. water-extraction preparation and ethanol-extraction preparation of three clear hypoglycemic compound strongly inhibitedα-glucosidase activity, showed marked concentration-effect relationship. water-extraction preparation was stronger than ethanol-extraction preparation,when its concentration of 25mg/ml, the equivalent concentration of acarbose concentration of 2.5mg/ml, the inhibitory rate reached 60%.2. Low dose group of three clear hypoglycemic compound had better lower fasting blood sugar and blood glucose after 2hr of streptozotocin induced rat model of type 2 diabetes , and the hypoglycemic effect was better than acarbose.3. Combined with in vitro and in vivo experimental results, it can be argued that inhibit the activity of a-glycosidase enzymes were identified is one of three clear hypoglycemic compound glucose-lowering mechanism.
