ObjectiveObserving the expression of Pre-stimulation with moxibustion on CDK5 and P27kipl in rats of Alzheimer's disease, and to research and discuss the mechanism of moxibustion AD prevention and treatment.MethodsAll the rats are randomly divided into four groups, for the pre-moxibustion group, rats were moxibustion Baihui and Shenshu points for eight weeks, and then in rats bilateral hippocampus are injected Aβ1-42 to establish AD model of rats. Through the orientation navigation experiment and space exploration experiment of the Morris water maze to test behavior of AD rats. Immunohistochemical method is used to test the expression of CDK5 and P27kipl in rat hippocampus organization.Results1. Through the behavioral detection of the Morris water maze Water maze:the index data, average escape incubation period in the orientation navigation experiment before five days and the numbers through the original escape platform and effective time on the sixth day, show that the sham operation group compared with the normal group has no obvious difference (P>0.05); however the model group compared with the normal group and the sham operation group is have significant difference (P<0.01); the group of advance moxibustion rats, memory and learning ability, is obviously superior than the model group(P<0.01).2. Observing the expression of CDK5 and P27kipl in rat hippocampus organization in each group by Immunohistochemical method:the normal group compared with the sham operation group, the expression of CDK5 and P27kipl has no obvious difference (P>0.05); but the expression of CDK5 in model group is significantly increased, while the expression of P27kipl is decreased accordingly, compared with before two groups, is have significant difference (P<0.01); the expression of CDK5 in the group of advance moxibustion rats is significantly reduced compared with the model group, the expression of P27kipl is increased obviously, there is a significant difference (P<0.01).Conclusions1. Kidney essence and insufficiency of marrow-sea are the main cause of senile dementia. For this cause, acupuncture to fill kidney essence the brain intelligence for main therapeutic principles. Baihui and Shenshu can be used as the main points of the disease prevention and treatment.2. moxibustion beforehand stimulation can effectively improve the learning and memory in AD model of rats, which is one way to effectively prevent AD, and also easy to learn and understand, can be widely used in clinical.3. The mechanism for Moxibustion to control AD is likely to decrease the expression of CDK5, increase the expression of P27kip1, extend cells life and play the protective role of nerve factors to inhibit AD pathologic features.
