The optimized values of every factors were determined, those to be glucose 24.34 g/L, peptone 22.14 g/L, urea 7.25 g/L, potassium dihydrogen phosphate 0.2 g/L, sodium chloride 0.005 mg/L, calcium chloride 0.01 mg/L, vitamin B1 100 mg/L, magnesium sulfate 0.3 g/Land a maximum dry weight of mycelium of 2.9082 g/L was obtained.Then, the formula of culture medium was optimized on the basis of, respectively, remarkable to the influence of several shake flask fermentation condition of. The optimum culture conditions was as follow is initial pH 5.6, culture temperature 26.6℃, incubation time 5.72 days, shaking speed 156.6 r/min, age of inoculum 4 d, inoculum level 7%, liquid volume 100mL/250 mL, and the best is 14.12 g/L. The validation experiments with the optimum medium and culture conditions were implemented in triplicate, the error between true value and predictive value is 1.13%.Finally, using univariate and response surface method of Cordyceps gunnii intracellular polysaccharide by ultrasonic extraction process was optimized. The optimum ultrasound extraction conditions were confirmed as follows: ultrasound extracting time 214.4 s, ultrasound power 413.7 W, the ratio of water to sample 46.9:1(mL:g). The predicted max extraction of polysaccharides is 2.2262%, Relative error is 1.8% with verification test extraction. And study its anti-tumor activity was investigated polysaccharides from Cordyceps gunnii source of various human tumor cell proliferations in vitro.
