Objective:Hepatocellular carcinoma (HCC) is one of the most frequent malignant tumors, and it is characterized by its high capacity for invasion and metastasis.Hepatocellular carcinoma is a major cause of cancer death, but the molecular mechanism for its development beyond its initiation has not been well clear. Significance of signal transduction and activators of transcription-3(STAT3) has been detected in a wide variety of human tumor cell including Hepatocellular carcinoma.The expression of matrix metalloproteinase-2(MMP-2) has been linked with tumor invasion, angiogenesis, and metastasis. It has been reported that STAT3 can induce MMP-2 expression in some cancer cells. However, the molecular basis for STAT3 regulates MMP-2 expression in Hepatocellular carcinoma remains unclear.The aim of our study is to explore the expression and clinical prognostic significance of STAT3 and MMP-2 in the hepatocellular carcinomas tissue and the relationship between the expression of STAT3 that in tumor tissues and MMP-2 in paraneoplastic tissues of HCC.To explore the expression and clinical significance of STAT3 and MMP-2 in the hepatocellular carcinomas tissue (HCCs). Methods:56 cases of hepatocellular carcinomas who had undergone surgery for HCC who had not been other treated, to detected expression of STAT3 and MMP-2 proteins of their tumor tissues and their paraneoplastic tissue by immunohistochemistry technique (SP),and 14 cases of normal liver tissue as control specimens. To analyzed the data with tumor pathological differentiation,survival and clinical feature.Results:The positive expression intensity of STAT3 in HCC was remarkably stronger than that abnormality liver tissues. The expression intensity of STAT3 and MMP-2 in HCC tumor tissues was 71.4%,73.2%, respectively.The expression intensity of STAT3 and MMP-2 in paraneoplastic tissues of HCC was 57.1%,80.4%, respectively. They were related to tumor pathological differentiation,survival and clinical feature.Moreover, there was a positive relationship between the expression of STAT3 that in tumor tissues and MMP-2 in paraneoplastic tissues of HCC. Expression STAT3 and MMP-2 protein have been closely related to a shortened survival.In both monovariate Kaplan—Meir plot and multivariate Cox regression analysis the expression of STAT3 protein and MMP-2 protein were linked to unfavorable prognosis. The hazard ratios of expression of STAT3 protein and MMP-2 protein were 2.250 and 1.513.Conclusions:The overexpression of STAT3,MMP-2 take part in HCC occurr and developing. respectively STAT3 and MMP-2 can be used as prognostic indicators after the operation..
