Objective: To study the distribution of interleukin-18 ( IL-18) gene promoter sequence -137C/G,the single nucleotide polymorphism genotypes and allele frequencies in Guangxi-guibei Hans population of China,and to analyze the association with type 2 diabetic nephropathy.Methods : Polymerase chain reaction-restriction fragment length Polymorphism (PCR-RFLP) analysis was used to detect IL-18 gene-137C / G polymorphism in Hans population in Guangxi-guibei region. The subjucts were 58 unrelated patients with diabetes mellitus (DM group) , 50 diabetic nephropathy (DN group) and 40 normal controls. At the same time, plasma lipids, lipoprotein levels, including serum total cholesterol (TC), triglyceride (TG), high density lipoprotein - cholesterol (HDL-C), low density lipoprotein - cholesterol (LDL-C), apolipoprotein A1 (apoAI), apolipoprotein B (apoB) was measured using automatic biochemical tests.Results: IL-18 gene -137C/G locus CC, GC, GG genotypes present in Hans population of Guangxi-guibei. The CC genetypes and C allele frequency distributions of SNP-137 were higher in DN patients without DN than that in healthy controls(P<0.05,OR=1.635,95%CI:0.978~2.732). The distribution of genetypes and allele frequencies of SNP-137 polymorphism had statistical difference between T2DM patients with DN. Diabetic nephropathy patients with TC, TG and LDL-C level was significantly different (P <0.05).Conclusions: There is IL-18-137C/G single nucleotide site polymorphism in Hans population of Guangxi-guibei. Its distribution are obvious differences among different ethnic groups. IL-18 gene -137 G/C polymorphism and diabetic nephropathy was associated, C allele may be a genetic risk factor incidence of DN. The polymorphism may be a high degree by promoting the expression of IL-18 increased risk of DN.
