This experiment investigated the methods to extract the alkaloids from Cassytha filiformis and addressed the priliminary analysis of its main chemical components. In addition,the experiment also examined its effects on the mouse hepatoma H22 cell in vivo and in vitro, and investigated the mechanism of action.Alkaloids were purified by the procedure of solvent extraction from Cassytha filiformis.The contents of it were determined by High Performance Liquid Chromatography-Mass Spectra(HPLC-MS).The inhibition effects of alkaloids from Cassytha filiformis on H22 cell in vitro were detected by Trypan blue staining and MTT assay.Ascites tumor model and solid tumor model of implanted H22 cell were bulit up to observe the effect of alkaloids from Cassytha filiformis on mouse hepatoma H22 in vivo.Besides, the spleen index and thymus index were detected to evaluate the toxicity and side effects of the alkaloids from Cassytha filiformis. Change in cell morphology in the cell H22 solid tumor of mice were observed by normal Hematoxylin-Eosin(HE) staining.DNA agarose electrophoresis was used to detect the characteristic changes of apotosis and the activity of DNA TOPO I.The effect of alkaloids from Cassytha filiformis on the exrpession of Bax and Bcl-2 of H22 cell were determined by using immunohistochemistry.The results indicated that the main content of alkaloids from Cassytha filiformis were actinodaphnine, neolitsine and dicentrine.It significantly inhibited the growth of H22 cell in vitro with a time-and dose-dependent releationship.Alkaloid from Cassytha filiformis could prolong the life span of the mice implanted in ascites with H22 cell in a cetern extent. Significant inhibition of the growth of H22 solid tumor was observed, through there were obvious toxicity and side effects were observed in the high-dose group.The alkaloids from Cassytha filiformis can also inhibit the activity of TOPO I.
