| Objective:To explore the correlation of the expression between Gas-17 and TFFs in gastric cancer and the effects of gastrin on cell proliferation and the expression of in gastric cancer cells.Methods:The expressions of Gas-17,TFF1,TFF2 and TFF3 were determined by immunohistochemical method in 120 gastric specimens including 30 cases of normal gastric mucosa,30 cases of intestinal metaplasia,30 cases of dysplasia and 30 cases of gastric carcinomas.MKN45 cells were incubated in the medium with Gas-17 (1~1000nmol/L), proglumide (1~10mmol/L) and the combination of these two agents (100 nmol/L Gas-17 and 1~10mmol/L proglumide).Cell growth and proliferation of MKN45 were analyzed with MTT assay.The expression of TFFs was determined by Western-Blot in MKN45 cells and were incubated with Gas-17 (1~100nmol/L), proglumide (10 mmol /L) while the combination of these two agents (100nmol/L Gas-17 and 10mmol/L proglumide).Results:In contral group with normal gastric mucosa,precancerous group and gastri carcinoma group, the expression levels of TFF1,FF2 had a decreasing tendency (p<0.05), the expression levels of TFF3,Gas-17 had a increasing tendency (p<0.05).Correlation analysis showed that there was a negative correlation between the expression of Gas-17 and TFF1,TFF2,while a positive correlation between the expression of Gas-17 and TFF3.The expressions of TFF1,TFF2 were significantly related with differentiation degree (p<0.05), but not with age,sex and stage The expressions of TFF3 were significantly related with differentiation degree and stage (p<0.05), but not with age,sex. MTT showed Gas-17 significantly promote cell proliferation at the concertration of l~1000nmol/L (p <0.05),PGL at the concertration of 1~10mmol/L significantly inhibite the proliferation of MKN45 cells (p<0.05). In the combination of these two agents, PGL (1~10mmol/L) significantly blocked and inhibite the proliferation of MKN-45 cells induced by Gas-17 (p<0.05). Meanwhile, WB showed gastric cancer line MKN45 have the expression of TFFs protein. Gas-17 at the concertration of 1~100nmol/L significantly strengthen the expression of TFF3 in MKN45 cells (p<0.05) and inhibite the expression of TFF1,TFF2. In the combination of these two agents, PGL significantly can inhibite the expression of TFFs stimulated by Gas-17 (p<0.05).Conclusions:There was a negative correlation between the expression of Gas-17 and TFF1,TFF2 in gastric cancer tissues while a positive relationship between the expression of Gas-17 and TFF3. Gas-17 would promote cell proliferation in human gastric cancer line MKN45 and inhibite the expression of TFF1,TFF2, strengthen the expression of TFF3. Its antagonist PGL significantly blocked and inhibite the role.The expression of TFF1,TFF2 were down-regulated and TFF3 was up-regulated may be the reason of Why Gas-17 play a key role in the development of gastric carcinoma. |
