Objective:To observe the effect of brain-derived neurotrophic factor (BDNF) on transplantation treatment of bone mesenchymal stem cells (MSCs) in rats of intracerebral hemorrhage (ICH).Methods:The intracerebral hemorrhage model of rats was established, and was injected the purified MSCs of cultured and marked in vitro through the left ventricle while injected BDNF locally. The each extent of neurological deficits score was evaluated after 7ds,14ds and 21ds. The MSCs migration and differentiation were measured by immunohistochemistry methods.Results:The dysfunction were serious in control and BDNF group by crawling measured, and the scores of them at each time point were higher than MSCs group and MSCs+BDNF group, furthermore, the scores were significantly increased in BDNF group at 14ds,21ds (P<0.01)compare with control group. The dysfunction were not serious in MSCs group and MSCs+BDNF group, and the scores were significantly increased in MSCs+BDNF group at 7ds,14ds (P<0.01) compare with MSCs group. The scores in each group were the highest at 7ds, decreased at 14ds, and significantly decreased at 21ds. It was significant (P<0.01) compare with each time, but no MSCs+BDNF group at 14ds and 21ds. Marked MSCs in vitro could remove the area around hemorrhage, the MSCs was find in nucleus marked by red BrdU, the positive cells of MSCs+BDNF group was more than MSCs group at each time (P<0.01).It showed that the MSCs around hemorrhage could differentiate into NEUN,GFAP and CNP cells by double fluorescence analysis. The positive cells increased endlessly follw time, and immunoreactive cells on NEUN,GFAP and CNP were increased of MSCs+BDNF group at 7ds,14ds and 21ds (P<0.01)compare with MSCs group. Conclusion:Using MSCs or BDNF lonely has protective effect in intracerebral hemorrhage reperfusion injury, but the protective effect of MSCs was obviously if using BDNF simultaneously, the survival time of MSCs around intracerebral hemorrhage was prolonged by BDNF, at the same time, the MSCs to NEUN,GFAP,CNP cells were promoted, and has a synergistic effect in this repairing process.
