Objective: To study the protection of the total flavones of propolis (TFP) on damage of intestinal mucosa by x-ray radiation in rats.Methods:50 SD male rats were randomly divided into 5 groups : Fifty rats were randomly divided into five groups : the normal group,the radiation group,the total flavone of propolis group(200mg/kg/d,100mg/kg/d),NAC group. The last four groups were irradiated in a single dose of 9Gy high-energy X-ray of the whole abdomen regions on the fourth day after intragastric administration. Rats were killed three days after irradiation. The small intestinal samples were collected for pathology analysis and examination of SOD activity and the contents of MDA and NO and MPO,and observation of apoptosis of mucosa cell by means of Tunnel. The blood samples were for examination of the content of DAO. The expressions of P38MAPK and ICAM-1 were assessed by using western blotting and immunohistochemistry respectively.Results:The model of radiation injury of intestine was built successfully. In the radiation group the mucosa structure of small intestine was damaged severely,the content of NO and MDA and MPO and the plasma lever of DAO were increased significantly (P <0.01). The activity of SOD were decreased significantly (P<0.05) compared with the normal control group. The apoptotic index were increased significantly (P <0. 05) Compared with the radiation group. Moreover,the expressions of P38MAPK and ICAM-1 increased significantly. The mucosa structure of small intestine in TFP group were improved greatly. The total flavones of propolis decreased the content of NO and MDA and MPO and the plasma of DAO,and enhanced the activity of SOD significantly (P<0.05). The total flavones of propolis could decrease Intestinal mucosa apoptosis and the expressions of P38MAPK and ICAM-1 in evidence.Conclusions:1. The total flavones of propolis can improve the SOD activity, reduce lipid peroxidation caused by oxygen free radical, inhibit the expression of MDA and radiation injury of apoptosis of bowel mucosa. Thus,it can improve the radiation damage of bowel mucosa in some degree, reduce radiotherapy side effects,enhance radiation tolerability.2. The total flavones of propolis can inhibit express of P38MAPK activation,reduce production of ICAM-1 and MPO, which indicate blocking intracellular signalling pathways may be a new way of protect the radioactive injury of the small intestine.
