Objectives:The objective of this study was to globally characterize the expression profile of transitional cell carcinoma by shotgun proteome strategy. On the basis of expression Profile, the tumor-stroma interaction and biomarker panel discovery from urine was discussed.Methods:Prospective collected specimens of four bladder transitional cell carcinoma. highly purify transitional cell carcinoma and stroma by laser capture microdissection (LCM); separation of peptide mixture by two-dimensional liquid chromatography (2D-LC) followed by electrospray ionization-tandem mass spectrometry (ESI-MS/MS) identification; bioinformatics analysis of the identified proteins; compare with the published literature.Results:A total of 775 proteins were commonly appeared in 4 specimens.612 of the 775 proteins with biological process annotation as to the Gene Ontology (GO). There were 40/25 GO terms exhibited as enriched/depleted within the biological process annotation. The enriched GO troms centered on metabolism process.117 proteins expressed both in cancer cells and stroma. Among them,73 proteins located in well-known pathways. Proteins belong to the term of cell adhesion, cell proliferation, cell differentiation are good candidate biomarkers for cancer detection from urine.Conclusions:The present expression profile providing valuable information for the understanding of cell biology, tumor-stroma interaction as well as discovery of biomarkers from urine.
