OBJECTIVE:To observe the effect of hyperuricemia on expression of Ki-67 and URAT1 of rat's aortic smooth muscle cells(ASMC). METHODS:20 male OLETF rats of 4 weeks age were raised up to 25 weeks when the rats were suffering from hyperinsulinemia,disturbance of lipid metabolism and adiposity, then were divided randomly into control group (n=10)and high purine diet group (n=10).The high purine diet group was administered with adenine (50mg/kg/d) by gavage and was fed ground standard rat chow containing 10% yeast.20 male LETO rats of 4 weeks age were processed the same way with OLETF.TG,TC,UA,CRE,BUN and 24h urine volume and UUA were detected. Insulin were detected by radioimmunity. The incidence rate of hyperuricemia was compared between the OLETF and LETO rats. Kill all of the rats after three weeks of high purine diet. The samples of thoracic aorta from rats were sliced into the paraffin sections. HE staining was used to observe the aortic smooth muscle structure and immunohistochemistry to detect the expression of Ki-67 and URAT1. RESULTS:1.①On the 11th day of high purine diet, blood uric acid level of both OLTEF and LETO high purine diet group were significantly higher than their corresponding control group (P=0.000). The hyperuricemia model of rats was formed successfully.;②The incidence rate of hyeruricemia of OLTEF high purine diet group (80%) was significantly higher than that of LETO high purine diet group(30%)(P<0.05).Moreover, blood uric acid level of OLTEF high purine diet group was much higher than that of LETO high purine diet group (P=0.00).That was to say the OLETF rats suffering from Reaven syndrome were more likely to develop hyperuricemia;③All of the rats showed no obvious signs of thoracic aortic atherosclerosis nor foam cells, so there were no statisticl significance between LETO and OLETF rats (P>0.05).④. No expression of Ki-67 and URAT1 was showed that there were also no statisticl significance between the two groups (P>0.05).⑤The expression of URAT1 was detected on fat cells. CONCLUSION:①High purine diet persisted for 3 weeks had no obvious effect on the expression of Ki-67 and URAT1 on 28 weeks old male rats, either LETO or OLETF, nor result in atherosclerosis. As one of the factors of atherosclerosis, it need more time for hyperuricemia to result in atherosclerosis.②Fat cell expresses URAT1 and that maybe have relevance with hyperlipemia,fat and hyperinsulinism etcl, but it should be further attested through more study.
