Study On The Chemical Substance Of Tranditional Chinese Medicine Of Gardeniae Fructus

In this paper, chemical substance of tranditional Chinese medicine of Gardeniae Fructus (GF) was studied, and a comprehensive quality standard was established. Details are as follows:Three-wavelength fingerprint of GF was established. LC-MS technique was applied for qualitative analysis, and thirteen compounds were identified by combing with related literatures; Five components of Genipin-1-gentiobioside, Geniposide, Chlorogenic acid, CrocinⅠand CrocinⅡin GF were determined; Quantitative analysis of multi-components by internal reference (QAMIR) was applied in FG quality evaluation, System suitability of a internal standard to determine different kind compounds was test. Geniposide, crocinⅠcould be the internal reference to determine Geniposide, crocinⅠ,Ⅱ, respectively. System suitability results of correction factor (f) were good (RSD<5%) and results of QAMIR and external standard method were of no significant difference (RSD<5%).Fingerprint and five main components contents of different processed drugs and different parts of GF were studied comparatively. Result showed that Chao Gardeniae Fructus (CGF) had little difference from GF while Jiao Gardeniae Fructus (JGF) distinguished much. Specifically, JGF had less chemical compositions and their main components contents were significantly reduced. While the result of whole GF, peel and seed research showed that fingerprint and main components contents varied greatly between the peel and the seed.Quality standards of GF were established. TLC was studied to identify Genipin-1-gentiobioside, Geniposide and CrocinI in GF simultaneously. Contents of moisture, ash, acid insoluble ash, alcohol extract of 28 batches GF were determined.95% confidence interval was calculated,80% and 120% confidence interval were taken as the limit standard, they were suggested not exceed 7.9%,7.6%,0.2%, while alcohol extract not be less than 9.1%. Three-wavelength fingerprint was established to show whole chemical characterizes of GF. Then, contents of Genipin-1-gentiobioside, Geniposide, Chlorogenic acid, CrocinⅠand CrocinⅡwere determined, as the same case, they should not be less than 6.0mg·g-1, 30.1mg·g-1,0.8mg·g-1/4.4mg·g-1,0.7 mg·g-1;...
