Viticis Fructus (Mangjingzi in chinese name) is a traditional chinese medicine prepared from the fruit of Vitex trifolia L. It has been recorded in Chinese Pharmacopoeia as a folk medicine. According to the theory of chinese medicine, Viticis Fructus has the funtion of Dispersing wind-heat and Removing heat for treating head and ocular disease, which has been used in the treatment of headache, cold, cancer, migraine, eyepain and rheumatism. For future reasonable development and application, we have systematically studied on the chemical constituents and determination methods of Vicitis Fructus. Our research results are as follows.1. Literature reviewAccording to the study purpose, conducted a literature review relating to the source, chemical constituents, pharmacological effect, Processing Technique, quality of Vicistis Fructus.2. Study on chemical constituents in fruits of Viticis Fructus.In this thesis, the chemical constituents of the extracts with 95% ethanol from Viticis Fructus were studied. 12 compounds were isolated with slica gel, high-speed counter-current chromatography, etc. On the basis of analysis NMR, MS and measuring of physic-chemical characters, 11 chemical structure were elucidated as casticin(Ⅰ), artemetin(Ⅱ),5,7,3,4-tetrahydroxy-flavone6-C-β-D-glucopyranoside(Ⅲ), 3,4-dihydrox-ybenzoic(Ⅳ), 4-hydroxybenzoic(Ⅴ), 3,6,7-trimethylquercetagetin(Ⅵ), ferulaldehyde(Ⅶ),β-stiosterol(Ⅷ), daucosterol(Ⅸ), vitexilactone(Ⅹ), vanillin(Ⅸ). Of all these compounds,Ⅲwas isolated for the first time from Viticis Frutus.3. Simultaneous quantitative analysis of four chemical compounds in Viticis Fructus by multi-components assay by single marker. In order to establish a new method of quality evaluation and validate its feasibilities by the simultaneous quantitative assay in Viticis Frutus, the new qulity evaluation method, quantitative analysis of multi-components by single marker(QAMS), was established and validated with Viticis Fructus. We used 3,4-dihydroxybenzoic acid, 4-hydroxybenzoic acid, 3,6,7-trimethylquercetagetin, casticin as analytes to evaluate the quality. We also determined 10 batches of samples, collected from different drug markets, by both external standard method and QAMS and found no significant differences between external standard method and QAMS. Finally we sure QAMS is feasible and suitable to evaluate the quality of Viticis Fructus.During the progress, we also used multi-index breakdown formula evaluation and orthogonal design to study the homogena extration techniques for Viticis Frutus which take 3,4-Dihydroxybenzoic acid, 4-Hydroxybenzoic acid, 3,6,7-trimethylquercetagetin, casticin as indexes. Finally, the optimum extraction procedure was extracting one minutes using 5 times volumes of 85% ethanol. Compared with other extaction measures, such as Reflux and Ultrasonic wave extraction, The homogena extraction technique was simple, quick and practical.
