The South Lake water chestnutis a kind of special aquatic plant produced in Zhejiang Jiaxing south lake.. According to traditional Chinese medicine dictionary, the shell of water chestnut can help to clear heart, refreshing, ease thisrty.the treatment of stomach, skin warts esophageal cancer, cervical cancer etc. It has been widely used in folk since hundreds of years. Survey shows gastric cancer is one of the most common digestive tract malignant tumors. Now in the comprehensive treatment of cancer, natural medicine treatment becomes increasingly important. So, deepening the plant active ingredient research is of important significance. The research first established the bionic extraction method according to semi-bionic extraction theory, taking inhibitory activity as bioactive guidance; then used uniform designation to optimize the design method, and studied the extraction time, material liquid ratio, dosage of pepsin, trypsin factors. The results showed that when lg shell powder was added to 10mL solvent (ratio of 1:10 of powder to solvent)in the absence of pepsin, extracted for 30 min at 37℃, and then centrifuged to collect precipitate, added lOmL solvent and 4% trypsin, extracted for 30 min at 37℃.the extracts had the highest activity against gastric cancer and the cell viability was the lowest, i.e.45.52%. Secondly, my research studied the functional ingredients of extracts. The results showed that the content of total flavonoids is 0.335±0.010 mg/mL,total polyphenols 3.747±0.038 mg/mL,total polysaccharide 0.910±0.001 mg/mL,total saponins 0.719±0.007 mg/mL; their oxidation resistance were analysed, including ABTS 4.118±0.006mg/mL AEAC. restore force 0.333±0.033mg/mL AEAC, FRAP 2.464±0.239 mg/mL AEAC. Furthermore, the repair of oxidative damage to HUVEC cells was examined. The oxidative damage model was built and then intracellular accumulation of lipid peroxides and antioxidant enzymes was investigated. The research updated the new semi-bionic extraction method and studied the bioactive ingredients, repair of oxidative damage to verify the bioactivity of water chestnut extract. The result showed that bioactive ingredients could reduce MDA content and protect SOD activity significantly. The bioactive ingredients from water chestnut have special usage in kinds of diseases. Further mechanism study on extraction, seperation, purification and function will promote the development of application in food and medical industry.
