Objective:To observe the effect of anticoagulant therapy on acute lung injury caused by paraquat poisoning.Methods:All 120 adult healthy male Wistar rats were randomizedly divided into three groups:the paraquat poisoning groups, rats were intragastric administration paraquat 50mg/kg body weight once at the beginning; anticoagulant therapy groups, rats were intragastric administration paraquat 50mg/kg body weight once at the beginning, then subcutaneous injections low molecular heparin calcium,68IU/60kg, twice a day, and intragastric administration aspirin,1.67mg/kg, once a day; the control groups, intragastric administration physiological saline. At the 3d, 7d,14d and 21d, rats were sacrificed postanesthetic respectively after paraquat exposure, sample of lung tissue and venous blood were collected. Prothrombin time, activated partial thromboplastin time,fibrinogen,thrombin time,D-dimer,lung tissue hydroxyproline were examined. Part of lung tissue were collected to observe by optical microscope, HE and Masson staining.Results:The hydroxyproline content of lung tissues, both of poisoning and anticoagulation therapy groups are higher than that of control groups at 3d,7d,14d and 21d (P<0.05)。The content of hydroxyproline of anticoagulation therapy groups are lower than that of paraquat groups at 7d,14d and 21d (P<0.05)。The PT,APTT,Fib,D-dimer, both of poisoning and anticoagulation therapy groups are higher than that of control groups at 3d (P<0.05). The D-dimer of anticoagulation therapy groups are lower than that of control groups at 3d (P<0.05)。The TT and D-dimer, both of poisoning and anticoagulation therapy groups are higher than that of control groups at 7d,14d and 21d (P<0.05)。The D-dimer of anticoagulation therapy groups are lower than that of control groups at 7d,14d and 21d (P<0.05)。The lung injury was gradually aggravate following the time going on, fibrosis locally,the therapy group changes slightly.Conclusion:Anticoagulation therapy can improve hypercoagulabale state caused by paraquat poisoning and decrease paraquat lung injury.
