Objective:To study the expression and function of chemokine receptor CXCR4 in human neuroblastoma cell line with high and low metastatic potential.Methods:Chemokine receptor CXCR4 mRNA was detected by RT-PCR, The expression of CXCR4 in neuroblastoma cell line was evaluated by flow cytometry; migration response to CXCR4 particular ligand-CXCL12 detected by chemotaxis assay.Results:Chemokine receptor CXCR4 was highly expressed on high metastatic potential cells, the relative amounts of CXCR4 mRNA were 0.57±0.08 and 0.29±0.05 (P=0.005<0.05). and the expression level of CXCR4 protein on cell surface were 64.59%±1.57% and 36.72%±0.57%(P=0.00<0.05). The specific ligand of CXCR4-CXCL12, induced migratory response of high metastatic potential cells(P<0.05).The appropriate response density was 1 OOng/ml. AMD3100 can inhibit the reflect(P<0.05).Conclusion:CXCR4 Was functionally and highly expressed on high metastatic potential cells and the interaction of its ligand CXCL12 with CXCR4 was possibly associated with tumor metastasis potential in vitro.
