Objective To study the relationship between Thl/Th2 cytokines expression in serum and prolonged cardiac allografts survival time induced by donor liver non-parenchymal cells (NPC) in rats.Methods Heart transplantations were carried out using SD rats as donor and Wistar rats as the recipient.40 Wistar rats were randomly divided into two groups:rejection group and tolerance group (each group 20 rats). Rejection group were injected 1ml normal saline via tail vein and 7 days later, the hearts from SD rats were transplanted to their neck; tolerance group were injected NPC 2×10'(1ml) from SD rats via tail vein and 7 days later, the hearts from SD rats were transplanted to their neck. Mean survival time (MST), mixed lymphocyte reaction (MLR), serum expression level of Th1/Th2 cytokines: interleukin-2 (IL-2), interferon-γ(IFN-γ), interleukin-4(IL-4), interleukin-6(IL-6), interleukin-10(IL-10) were measured.Results (1) MST (30.83±2.55d) of heart allografts in tolerance group was significantly longer than that (8.15±2.08 d) in rejection group (P<0.01). (2) MLR showed stimulation index (SI) of tolerance group (3.97±0.33) was significantly lower than that in rejection group(7.09±0.25) when SD rats spleen cells were stimulating cells (t=17.00,P=0.00); the SI was no difference between the two groups (tolerance group:7.36±0.31,rejection group:7.51±0.70) when Lewis rat spleen cells were stimulating cells(t=0.45, P=0.67); (3) The serum expression level of cytokines before the operation was no difference between the two groups; 7 days and 14 days after transplantation, the serum expression level of IL-2, IFN-γ(Th1 cytokines) in rejection group were much higher than those in tolerance group, and serum expression level of IL-4, IL-10 (Th2 cytokines) in rejection group were much lower than those in tolerance group (all P<0.01); yet serum expression level of IL-6(Th2 cytokines) in rejection group were much higher than that in tolerance group (P<0.01) 7 days after transplantation.Conclusion 1 NPC can prolong cardiac allografts survival time in rat heterotopic heart transplantation.2 The effect induced by NPC is donor specific.3 The bias from Th1 cell reaction to Th2 cell reaction plays a pivotal role in prolonging cardiac allografts survival time induced by NPC.
