Objective:To study the mechanism of fine particulate matter(PM2.5) damaging endothelial cell from oxidative pathway. And to show the efficacy and mechanism of ginsenoside Rgl on the inhibition of endothelium injuries by exposure to PM2.5. Moreover, to reveal the variation of Nrf2and HO-1in the Nrf2-ARE signaling pathway from the molecular and gene levels.Methods:Human umbilical vein endothelial cells(HUVECs) as research object.Screening of suitable exposure concentration of PM2.5and the optimum concentration of ginsenoside Rgl.(1)Set control group and PM2.5treatment group (concentrations including0.1,0.2,0.4,0.8mg/ml).(2) set control group,PM2.5treatment group (0.8mg/ml),PM2.5and Rgl treatment group(0.8mg/m1+0.04mg/ml) and Rgl treatment group(0.04mg/ml). After treatment24h,each group was detected cell growth situation through MTT,intracellular reactive oxygen species (ROS) level through fluorescence labeling probe method and HO-1,Nrf-2mRNA expression level through RT-PCR method.Statistical processing among multiple groups with one-way ANOVA.Results:In certain range of exposure concentrations, there is decreased cell survival, elevated levels of intracellular oxidative stress,increased expression of HO-1mRNA and no statistical difference of Nrf2mRNA expression as the PM2.5exposure concentration increasing. The concentration of PM2.5respectively was0.1,0.2,0.4and0.8mg/ml.The OD value respectively was0.67±0.10,0.63±0.1,0.59±0.06(P<0.05) and0.48±0.07(P<0.01).The fluorescence gray value respectively was179±5,191±6.198±211±11(all were P<0.01).The average optical density ratio of HO-1respectively was0.718+0.06,0.737±0.052,0.737±0.009,0.806±0.037(P<0.01).The average optical density ratio of Nrf2respectively was0.941±0.074,0.991±0.064,0.996±0.040,1.010±0.039(all were P>.05). After administration of Rg1, cell survival increases, level of intracellular oxidative stress decreases significantly and expression of HO-1mRNA increases in PM2.5and Rgl common treatment group(0.8mg m1+0.04mg/ml),while expression of Nrf2mRNA has no statistical difference, compared with0.8mg/ml PM2.5group. The OD value was1.06±0.17(P<0.01), the fluorescence gray value was147±13(P<0.01),the average optical density ratio of HO-1was0.874±0.040(P<0.05) and the average optical density ratio of Nrf2was1.039±0.061(P>0.05).Conclusion:(1)Oxidative stress is one of the pathways in PM2.5damaging endothelial cell.(2)Rgl counteracts endothelium activation by inhibiting PM2.5damaging HUVECs and the protection mechanism is anti-oxidant.(3) The process of Nrf2mediating the expression of HO-1maybe post-translationally regulated.
