ObjectiveTo explore the influence of Dihydroartemisin on the activity of Caspase-3and the growth of human ovarian cancer cell line HO-8910transplanted tumor under the skin of nude mice.MethodTo establish the subcutaneously transplanted tumor of human ovarian carcinoma in nude mice and then the mice were randomly divided into low-dose-group of Dihydroartemisin, high-dose-group of Dihydroartemisin, saline group, DDP positive control group; five nude mice each group, the nude mice was weighted before and after taking medicine, the variational curves of weight was drawed, and then DDP group was continuously received intraperitoneal injection for10days, other three groups were continuously received gavage for10days; the nude mice was killed through cervical dislocation after24hours of discontinuation, the transplanted tumor was weighed and the inhabitation rate was calculated and Immunohistochemistry method was used to detect the positive rate of Caspase-3in each group.ResultThe average inhibition rates of the low and high-dose group of dihydroartemisin based groups and DDPgroup were30.00%,48.03%and60.06%respectively, which was statistically significant compared with the saline group, and low-dose-group of Dihydroartemisin compared with DDP group and high-dose-group of Dihydroartemisin was also statistically significant, while comparison between high-dose-group of Dihydroartemisin and DDP group has no statistical significance. When use Immunohistochemistry method to detect the positive expression rate of Caspase-3, The positive rates of saline group, the low and high-dose group of dihydroartemisin based groups and DDP group were18%,29%,73%and80%respectively, with the increase dose of Dihydroartemisin,the activity of Caspase-3was increased gradually, there is no statistical significance between high-dose-group of Dihydroartemisin and the DDP group,but statistically significant among other groups.ConclusionDihydroartemisin can inhibit the growth of tumor in human ovarian cancer cell line Ho-8910subcutaneously transplanted nude mice,maybe through activating Caspase-3and inducing cellapoptosis as ananti-tumor effectin vivo.
