The biomedical model is gradually changing to the eco-medical, with the advancing in modern medical researching and the development of science and technology. So, the traditional Chinese pharmacology has become increasingly concerned by domestic and foreign researchers.Recently, the traditional extraction methods of Chinese traditional medical herbs have some drawbacks, such as extraction time is too long, low yield, high impurities. These are restricting of the development of Chinese traditional medical herbs, but also limiting the internationalization of Chinese medicine products. Therefore, select the appropriate extraction methods and the purification are the prerequisite and the key for the study of Chinese traditional medical herbs.In this paper, we extract Eleutheroside B by the traditional ethanol, CO2 supercritical extraction and ultrasonic extraction. By single-factor experiments and orthogonal approach optimize the extraction process of Eleutheroside B, ultimately determine the optimum extraction conditions and extraction rate are:1,Traditional ethanol reflux:Optimum conditions are liquid ratio 1:20,65% ethanol, time 3 h, and extraction rate is 1.89%.2,CO2 supercritical extraction:Optimum conditions are extraction pressure 30 MPa, extraction temperature 40℃, extraction time 2.5 h, CO2 flow rate 20 L/h, extraction rate is 2.67%.3,Ultrasonic extraction method:the best extraction conditions are 20℃, liquid ratio 1:25, power 550 W, extraction time 2 h, extraction rate is 3.74%.Comparison of the three different extraction methods, although the ethanol extraction method is simple, but required a longer time and the extraction rate is low; The time of the CO2 supercritical extraction is shorter, the extraction rate is moderate, but because of the higher operating pressure, it does not apply to large-scale continuous extraction; For ultrasonic extraction, it not only the extraction time is short, also the energy consumption is lower, and the extraction rate is highest, so we select the ultrasonic extraction as the best experimental method of Eleutheroside B.Purification of extracts selects the macroporous resin purification, by comparing the five different models of the macroporous resin static adsorption rate, determine the best purification of Eleutheroside B is AB-8 macroporous adsorption resin. The best purification procedure is:sample sdution's concentration 10 mg/mL, sample solutiorvs spale velocity 2 SV, pH=6,2 BV,60% ethanol with 1.5 SV elution flow rate, the refined purity is 67.71%.Use the UV detection, infrared detection and HPLC detection to determine the substance of Eleutheroside B.This study show that:ultrasonic extraction and purification of macroporous resin for Eleutheroside B, can be used for large-scale production, it's significant for Chinese medicine towards modernization and enter the international market.
