Aim:Acute Myocardial Infarction(AMI) is a world-wild disease that endanger people's health, both the morbidity and the fatality is very high. This article aimed to investigate the protective effects of Eleutheroside E on ischemia heart after acute myocardial infarction induced by pituitrin.Methods: Acute myocardial infarction animal models were induced by caudal vein injection of pituitrin 20U/kg weight. Observe the electrocardiogram and myocardiac mechanics changes as time going after injection of pituitrin. With verapamil 1mg/kg weight as positive control group, EleutherosideE5mg/kg weight were used 20 minutes before or 1 hour after injection of pituitrin respectively. And to observe the effects of EleutherosideE on electrocardiogram and myocardiac mechanics in acute myocardial infarction rats, and to observe the effects of EleutherosideE on contents of superoxide dismutase(SOD),malonaldehyde(MDA),Lactic dehydrogenase isoenzyme1 (LDH1),creatine kinase isoenzymes MB(CK-MB),the ratio of glutamic-oxal(o)acetic transaminase /glutamate-pyruvate transaminase(AST/ALT)in serum and myocardium of acute myocardial infarction rats. In order to exclude the influence of nerve and humor on heart in vivo, we utilize Langendorff isolated heart perfusion system, to observe the effects of Eleutheroside E 100mg/L on myocardial infarction induced by pituitrin.Results:In vivo experiments, it suggested that after injection of pituitrin 20U/kg weight, 1. Electrocardiogram had a change as following: (1)S-T segment rose≥0.2mV,AMI models were made successfully.(2) T wave rose and achieved peak at 30s, then S-T segment and T wave began to decrease after 30s, but they are higher than normal all the time;2. Myocardiac mechanics had a dynamic change:(1) myocardiac mechanics index rose immediately after pituitrin injection, and achieve peak in 10 minutes, compared with control,mean arterial pressure (MAP) increased 22.02%,left ventricular systolic pressure(LVSP)increased 20.89%, and the absolute value of left ventricular pressure maximal change rate (|±dp/dtmax|) increased 25. 94 % and 28.41% respectively significantly(P<0.01);(2) 20 minutes later, MAP,LVSP,|±dp/dtmax| began to decrease, LVEDP rose higher, 1 hour after pituitrin injection, compared with control, MAP decreased 23.34%, LVSP decreased 25.67%, +dp/dtmax decreased 18.72% and |-dp/dtmax| decreased 20.20% significantly(P<0.01). (3) 2h after pituitrin injection, compared with control, MAP decreased 28.32%, LVSP decreased 22.27%, +dp/dtmax decreased 27.83% and |-dp/dtmax| decreased 38.41% significantly(P < 0.01) 3.The level change of biochemistry index in serum and myocardium of acute myocardial infarction rats induced...
