PART1PREPARATION OF THE LOADING-IL-8MONOCLONAL ANTIBODIES TARGETED ACOUSTIC CONTRAST AGENTObjective:The purpose of this study was to prepared a kind of Microbubbles targeting loading-IL8ultrasound contrast agent, detected their physical roperties.Methods:(1)Using SPDP crosslinking to prepared a kind of Microbubbles targeting loading-IL8ultrasound contrast agent, by IL-8monoclonal antibody andSonoVue microbubbles.Observe the physical properties and stability.(2)Identification of IL-8monoclonal antibody conjugated with acoustic microbubbles:Take the coupling of SonoVue microbubbles suspension and SonoVue microbubbles suspension,Then add to the secondary antibody to ordinary microscope observation of the agglutination reaction.Results:(1) The Microbubbles targeting loading-IL8ultrasound contrast agent were similarWith SonoVue microbubbles in size, shape, traits.But the concentration was slightly lower.(2)Microbubbles targeting loading-IL8ultrasound contrast agent Agglutinated,SonoVue microbubbles did not agglutinated.Conlusion:Prepared to be successful. The micro-bubble size, shape, character, did not significantly change as the role of contrast agents in vivo. Objective:Model of rabbit abdominal aorta atherosclerotic plaques, fot the next Experiment.Methods:Model of rabbit abdominal aorta atherosclerotic plaques,Using ultrasound and pathological observation of aortic atherosclerotic plaques in rabbit abdominal diagnosis.Results:Ultrasound showed the model of abdominal aortic many plaques. Plaques pathological findings:endometrium was significantly thicker, deep to the membrane have a large number of foam cells and form plaques, the proliferation of medial smooth muscle cells, disorganized, and for plaque formation.Conlusion:The atherosclerotic rabbit abdominal aorta after ultrasound and pathology confirmed the establishment of successful. Objective:To discuss the diagnostic value of loading-IL8microbubbles in atherosclerosis.Methods:Observation of imaging characteristics in rabbits by using SonoVue ultrasound contrast agent and loading-IL8microbubbles.Then Comparative analysis of the intima, atherosclerotic plaque video density by Photoshop7.0And analyzing these indicators of ultrasound mean, peak strength, imaging time, peak time, time to half value, and so on, by Qlab8.0offl-ine software.Results:17model rabbits, ultrasound examination showed plaques in19, low echo6place, isoechoic7place, high echo6place.Using SonoVue ultrasound contrast agent Showed22place of patches, for the2D was not detected in patches of3.A significantvisual increase could be detected in plaquesafterusing microbubbles(P <0.01). Comparison of targeted contrast with ordinary imaging, intimal and plaque ultrasound mean and peak intensity was significantly different (P<0.05) Significant difference in intima/lumen and plaque/lumen ratio(P<0.05). Significant difference was shown in comparing intima and plaque enhanced drop half time (P<0.05).Conlusion:Have a higher sensitivity, resolution of details more clearly.By Microbubbles targeting loading-IL8ultrasound contrast agent to Atheromatous Plaques of Abdominal Aorta in Rabbits.This laid the foundation for further study the practical application of targeted ultrasound contrast agent.
