ObjectiveRabbit animal surgical model was used to simulate the artificial joint replacement, occurrence of aseptic loosening of prosthesis, and prosthesis revision surgery. Drug treatment was applied to promote osteointegration around the prosthesis after revision surgery. We hope to provide experimental basis for drug treatment of aseptic loosening of prosthesis, promote prosthesis bone integration after revision surgery.Methods30male New Zealand white rabbits were randomly divided into three groups (normal group, the experimental group and control group)In each group10,Normal group prosthetic titanium rod implanted in the tibial cancellous bone area, the experimental and control groups were implanted prosthetic titanium rod and titanium particles after8weeks, three groups of the reunification of the prosthesis. Experimental group after8weeks of treatment with alendronate subjects, the control group and normal renovation of the group were drawn. Introduced through the prosthesis mechanical experiments, hard Histological examination, evaluation of alendronate renovation of periprosthetic bone integration after the impact.ResultProsthesis introduced mechanical experimental results show that the experimental group (alendronate treatment group) at the maximum launch power is significantly greater than the control group. Hard histological sections by the picric acid poinsettia stained periprosthetic bone mass in the experimental group no significant loss of the control group of periprosthetic bone less or even bone defects. The results showed that osteolysis renovated by Allen sodium medication to promote the integration of the bone after the renovation.ConclusionsThe biphosphate—alendronate can promote prosthesis osteointegration after wear particles induced osteolysis revision surgery.
