Objective:1To explore the role of oxidative stress in preeclampsia by investigatingserum values of lipid profiles (TG, CHOL, HDL, LDL, APOA and APOB) andlipid peroxidation (MDA,8-iso-PGF2α).2To analyze the correlation between lipid peroxidation and lipid profiles,and to further investigate the correlation between lipid peroxidation and theseverity of preeclampsia.Methods:60women with preeclampsia enrolled as preeclampsia group,which was divided into mild preeclampsia group (MPE) and severepreeclampsia group(SPE).30pairs of normal ones served as controls.Maternal peripheral blood specimens were collected for8-iso-PGF2αmeasurement by ELISA and MDA by thiobarbituric acid (TBA) colorimetricassay, and lipid profiles were assessed at the center of blood test in ourhospital. All the data were analyzed by SPSS13.0software.Results:1Serum MDA and8-iso-PGF2α values in the preeclampsia group were(4.92±1.61)nmol/ml and (22.23±3.62)ng/ml respectively and significantlyhigher than those in controls [(3.95±1.10)nmol/ml, P=0.006;(19.91±2.83)ng/ml, P=0.044)]. Compared with MPE group, the serum MDA(5.19±1.77) nmol/ml and8-iso-PGF2α (23.13±3.63)ng/ml values in SPE[(4.25±0.83)nmol/ml,(19.98±2.52)ng/ml] showed significant increase(P=0.025; P=0.009).2Serum TG values in preeclampsia group (3.82±1.15)mmol/L weresignificantly higher than those in controls (2.96±0.77)mmol/L (P=0.001).Compared with those of mild preeclampsia, the serum TG values in severepreeclampsia showed no significant difference (P>0.05). Serum HDL values in preeclampsia group (1.64±0.43)mmol/L were significantly lower than thosein controls (1.91±0.48)mmol/L (P=0.015). Compared with those of mildpreeclampsia, the serum HDL values in severe preeclampsia showedsignificant decrease (P=0.006).3Positive correlation was founded between MDA,8-iso-PGF2α valuesand the ascending of TG (r=0.460, P=0.011; r=0.567, P=0.001) in severepreeclampsia, while the negative correlation was founded between8-iso-PGF2α values and the levels of HDL (r=-0.382, P=0.037).4Positive correlation was founded between serum8-iso-PGF2α andMDA values, systolic pressure, diastolic pressure in severe preeclampsia(r=0.450, P=0.013; r=0.378, P=0.039; r=0.578, P=0.001). No correlation wasfounded between serum MDA values and systolic pressure, diastolic pressurein severe preeclampsia (P<0.05, respectively).Conclusions:1Serum values of MDA and8-iso-PGF2α rise significantly in gravidaswith preeclampsia suggesting oxidative stress may play an important role inpathophysiology.2Serum TG values were increased significantly in severe preeclampsiaand there was a positive correlation between serum TG values and the level oflipid oxidative stress. Serum HDL values were decreased significantly insevere preeclampsia and there was a negative correlation between serum TGvalues and the level of lipid oxidative stress. TG and HDL values may reflectthe extent of oxidative damage.3Serum HDL and the level of lipid oxidative stress showed significantlydifference in mild preeclampsia and severe preeclampsia. This indicated thatlipid profiles and the level of lipid oxidative stress may contribute tomonitoring the severity of preeclampsia. 4The index of8-iso-PGF2α may be more precise in reflecting the levelof oxidative stress than MDA.
