Objective: To explant culture primary glioma cells and testchemotherapy drugs sensitivity in vitro, study the differences of drug typesand concentrations in the inhibition effect on cell proliferation.Method: Culture the primary glioma cells of38diagnosed as glioma andunderwent surgical removal patients from2010.6to2011.1in The SecondHospital of Hebei Medical University. These cells were grouped in randomand cultured with carboplatin, fotemustine, temozolomide and vincristine. Thecells activity were tested by MTT assay and flow cytometry, results werestatistically analyzed.Results: Carboplatin, fotemustine, temozolomide and vincristine were allexhibited the different anti-tumor effects, the sequence inhibit rates to these38cases glioma cells were26.32%,34.21%,44.74%,15.79%. The positiveexpression of O~6-methylguanine-DNA methyltransferase shows resistanceto alkylating agents chemotherapy drugs. High grades glioma cells alsomanifested the more notable resistance. The4drugs' sequence inhibit rates toastrocytoma were54.55%,54.55%,72.73%,36.37, and which to glioblastomawere10%,15%,30%,5%.The difference between them was statisticallysignificantly. The sequence proliferation index of the4drugs tested by flowcytometry were40%,42%,37%,51%, the dates were adopted for comparisonbetween groups, shows statistically significantly difference except Carboplatinvs. fotemustine(P=0.074).Conclusion: Each chemotherapy drugs exhibited distinctive activityinhibition to glioma cells. In comparison, temozolomide and fotemustineshowed the more noticeable activity inhibition than Carboplatin andvincristine. The inhibition effect was higher by the increasing of drug'sconcentrations. Grade IV glioma cells had more chemotherapy drug resistance than other type. The positive expression of O~6-methylguanine-DNAmethyltransferase shows resistance to alkylating agents chemotherapy drugs.
