Objective:(1)To determine the expression of L-selectin, ligand SLeXand FUT7synzyme in human normal liver cells lines L-02and liver cancer cell lines HepG2, andto determin the relevance between the changes and liver cancer migration.(2)Toresearch the impact of low molecular weight heparin (LMWH) to the expression ofL-selectin, SLeX, FUT7and adhesion and migration on HepG2cells(.3)To discuss thechanges of the migration and adhesion after suppressing the expression of L-selectinand ligand SLeX.Method:(1)The expression of L-selectin,SLeXand FUT7in humannormal liver cell lines L-02and liver cancer cell lines HepG2were examined byRT-PCR, Western-blot and immunohistochemistry.(2)The migration and adhesion ofHepG2cells were examined by Transwell and Fibronectin protein test.(3)Using MTTtest and RT-PCR, Flow Cytometry to detect the expression of L-selectin, SLeXandFUT7and the cell growth inhibition rate after the effects of LMWH(.4)Using LMWHto suppress the expression of L-selectin, using SLeXantibody to close down theexpression of SLeX, then take cells into24wells transwell room to detect the changesof HepG2cells migration, and the adhersion were examined by FN protein test. Result:(1)To the level of RNA, L-selectin, SLeXand FUT7was expressed in HepG2cellsbut not in L-02cells; to the level of protein,L-selectin expressed in HepG2cells was(10.09±0.5)%,the expression of SLeXwas high in HepG2cells but not in L-02cellsdetected by Western-blot and immunohistochemical staining.(2)As the ability ofLMWH increasing, the expression of L-selectin and FUT7mRNA were reduce(P<0.01),but have no effects on SLeX(P>0.01)。(3)The result of MTT methodshowed that LMWH have inhibition of growth to HepG2cell(sP<0.01)。(4)To Reducethe expression of L-selectin and SLeXcan lead the number of HepG2cell punching outmembrane reduced (P <0.01); the number of FN cell adhesion was reduced too.(P <0.01).Conclusions:(1) L-selectin,SLeXand FUT7were expressed in HepG2but notin L-02cells.(2)LMWH may inhibit the expression of L-slectin and FUT7and wouldinhibit the growth,migration,adherence of HepG2cells.
