Laryngeal carcinoma (LC) is one of the common malignant tumors in head and neck,the incidence of LC shows an upward tendency.The highest incidence of LC is in the northeastern region,and accounting for 5.7%~7.6% of the total malignancies.The invasion and metastasis of Laryngeal cancer ,which is a multi-step, multi-stage complex process.In this process,the malignant growth of tumor cells and its deprivation from primary tumor are the critical factors of tumor metastasis.The adhesion of tumor cell with endothelial cell,extracellular matrix and organ provids the molecular basis for the formation of metastatic lesion. During the adhesive process of tumor cell with endothelial cell and extracellular matrix,the combination of selectin with its specified ligand make a important role.The CD24 antigen,a low molecular weight and high glycosylated protein,through glycosylphosphatidylinositol anchors cell surface, it has been identified as a ligand of P-selectin, an adhesion receptor on activated endothelial cells and platelets. Overexpression of CD24 is in a variety of tumors, and it is relate with tumor invasion and metastasis.The SleX antigen, a carbohydrate contains oligosaccharide,it expresses on the glucolipide and glucoprotein of tumor cell surface. The SleX antigen has been identified as a ligand of E-selectin,which expresses on the cell surface of granulocyte and tumor cell mainly.The specific combination of SLeX with E-selectin assists cancer cell to permeate basement membrane of endothelial cell,and it shows a positive correlation with the aggravation and metastasis of malignant tumor.To study the CD24 and SLeX expression associated with laryngeal squamous cell carcinoma (LSCC), from the protein level to explore further the pathogenesis of LSCC,we detected different CD24/SleX expression of laryngeal mucosa lesions by immunohistochemistry.We choosed 42 case of LSCC and 24 case paracarcinous tissue ,20 case normal tissue.Experiment were divided into two,one group of HE staining diagnosed, another group SP immunohistochemical staining detected CD24/SLeX in those difference tissue.Standards were determined used a semi-quantitative analysis.Statistical analysis used chi-square test or Fisher's exact probability,the relation of two data used Spearman.In the study, positive expression rate of CD24 in laryngeal cancer tissue,adjacent tissuses and normal tissues was 64.29%, 33.33% and 25.00% respectively. Its strong positive expression rate was 35.71%,4.17% and 0.00% respectively. It showed that there was statistic significance (P<0.01) on difference among laryngeal cancer tissue,adjacent tissuses and normal tissues. Positive expression of CD24 in laryngeal cancer tissue was higher than that in adjacent tissuses and normal tissues. therefore, there was relativity between high expression of CD24 and occurrence of larygocarcinoma.Positive expression rate of SLeX in laryngeal cancer tissue,adjacent tissuses and normal tissues was 71.43%, 37.50% and 30.00% respectively. Its strong positive expression rate was 50.00%,8.33% and 5.00% respectively. It representated that the difference among laryngeal cancer tissue,adjacent tissuses and normal tissues showed significance (P<0.01)in statistics. Results indicated that the positive expression of SLeX in laryngeal cancer tissue was higher than that in adjacent tissuses and normal tissues, therefore, there was relativity between high expression of SLeX and occurrence of laryngeal carcinoma.Meanwhile, the study also showed that there was significance in statistics (P<0.05)on difference among strong positive expression of CD24/SLeX, metastases to lymphnodes of the neck, clinic stages and histopathology classification. It was obvious that strong positive expression of CD24/SLeX in those patients who had metastases to lymphnodes of the neck was higher than those patient who had not (P<0.05). In clinic stages, strong positive expression of larygeal patients at stage III-Ⅳwas higher than that of larygeal patients at stage I-II(P<0.05).In classification of histopathology, strong positive expression of larygeal patients with lower polarization was higher than that of larygeal patients with higher polarization(P<0.05). This result fulfiled with privous findings. According to research result, we assume that excessive expression of CD24/SLeX makes cancer cells infiltrates aroud tissues and speeds up occurrence, development and transfer of tumour.We also found that there was similar positive expression rate between CD24 and SLeX. The similar rate between them reached 64.29%. After processing by statistic manner, it can be found that there appeared positive correlation on expression level of CD24 and SLeX in LSCC tissue.It showed that cases with strong positive expression of CD24 and SLeX tended to infiltrate and transfer invasion and metastasis more.On above findings, expression rate of CD24/SLeX in normal tissues and adjacent tissuses was lower, whereas that of expression rate improved in laryngeal squamous cell carcinoma tissue. Therefore, there was relativity between high expression of CD24/SLeX and occurrence of laryngeal cancer. There was statistic significance on difference among strong positive expression of CD24/SLeX, metastases to lymphnodes of the neck, clinic stages and histopathology classification. Therefore, there was relativity between high expression of CD24/SLeX and invasion and matastasis of larygeal cancer , and positive rate of CD24/SLeX in laryngeal carcinoma showed significantly correlated obviously.In the study, it was firstly examined the expression of CD24 and SLeX together in laryngeal cancer tissues,adjacent tissuses and normal tissues. The result indicated that laryngeal cancer tissues was sensitive of the examination of CD24and SLeX, and there was statistic significance on difference among strong positive expression of CD24 and SLeX, metastases to lymphnodes of the neck, clinic stages and histopathology classification. It accounted for that examining expression of CD24 and SLeX in laryngeal cancer tissues could improve veracity in estimating transfer of laryngeal. Positive rate of CD24 and SLeX obviously appeared significantly correlated. Therefore, co-examining expression of CD24 and SLeX in laryngeal cancer tissues could reflect characteristics of clinic pathology in laryngeal cancer.In conclusion, research findings displayed that CD24 and SLeX involved in invasion and metastasis of tumor cells. Their expression in LSCC had relations with the invasion and metastasis of laryngeal cancer, and could reflect potential of the invasion and metastasis of laryngeal cancer.Therefore, we assumed that CD24 and SLeX could be selected as a symbol of tumour biology.By employing SP analysis to CD24 and SLeX in laryngeal tissue, we could ensure whether CAMs inhibitor could be applied or not. In addition, problems of the synthesis of CAMs inhibitor,and the detection and clinical application of the research need to be solved in further study.
