Objective:To explore the intervention effect of atorvastatin on mid-grade sepsis bydetecting the expression of IL-17,IL-10in peripheral blood and the expression ofIL-17in liver and kidney tissues of mid-grade septic rats.Method:1,Eighty healthy male S-D(Sprague-Dawley) rats of clean grade wererandomly divided into four groups: the normal group(A),the sham-operated group(B),the mid-grade sepsis group(C)and the Atorvastatin treatment group(D). Rats in A,B,C groups were treated with starch40mg/Kg/d for5days,while rats in D group weretreated with Atorvastatin40mg/Kg/d for5days. On the fifth day, Rats in Group Areceived no treatment,rats in Group B were sham-operated,while rats in group C andgroup D were adopted to make the mid-grade septic model by cecal ligation andpuncture(CLP);2,At0hours of the experiment,we collected blood from the tailvein.After72hours, we collected blood through carotid artery and removed the liver,kidney tissues from the abdominal cavity. To detect the expression of IL-17,IL-10inperipheral blood and the expression of IL-17in liver and kidney tissues of four groupsby enzyme-linked immunosorbent assay (ELISA)and immunohistochemistryrespectively.To observe the pathologic construction changes of liver and kidneytissues by HE staining,to determine the level of the aspartate aminotransferase(AST),alanine aminotransferase (ALT),blood urea nitrogen (BUN)and serum creatinine(Scr) in peripheral blood by automatic biochemical analyzer.Results:1,At0hours of the experiment,The IL-17,IL-10,AST,ALT,BUN,Scr level in the peripheral blood was similar in the four groups,there was nosignificant difference(P>0.05); At72hours of the experiment,The IL-17,IL-10,AST,ALT,BUN,Scr level in the peripheral blood was similar in the normal groupand the sham-operated group(P>0.05),Meanwhile the level of IL-17,AST,ALT, BUN,Scr was obviously higher in the mid-grade sepsis group compared with that ofthe Atorvastatin treatment group, the normal group and the sham-operatedgroup(P<0.05), the level of IL-10was significantly lower in the mid-grade sepsisgroup compared with that of the Atorvastatin treatment group(P<0.05) but was higherthan that of the normal group and the sham-operated group;2,In the liver and kidneytissues of the normal group and the sham-operated group, only a few scattered IL-17positive cells could be found, while in the mid-grade sepsis group, plenty of IL-17positive cells were present in the liver and kidney tissues.The quantity of IL-17positive cells in the Atorvastatin treatment group was higher than that in the normalgroup and the sham-operated group, but was significantly fewer than that of themid-grade sepsis group(P<0.05);3,From the pathological section of HE staining, wecould see severe inflammatory reaction and cell damage in the liver and kidneytissues of the mid-grade sepsis group,much lower inflammatory reaction and celldamage in the Atorvastatin treatment group than that of the the mid-grade sepsisgroup and no abnormality reaction in the normal group and the sham-operated group.Conclusion:1,There was a higher expression of IL-17in the peripheral blood,theliver and kidney tissues of the mid-grade septic rats, it may have reference value forthe incidence of the mid-grade sepsis;2,There was a lower content of IL-10inperipheral blood of rats in the mid-grade sepsis group compared with that of theAtorvastatin treatment group and the inflammatory response was more serious;3,Atorvastatin could regulate immunity and alleviate inflammatory response of themid-grade sepsis through down-regulation the expression of IL-17and up-regulationthe expression of IL-10;4,Atorvastatin may play a protective effect on the livertissues by lowering the expression of IL-17which is one kind of pro-inflammatorycytokines and reducing the extent of the inflammatory response and cell damage ofthe mid-grade sepsis;5,Atorvastatin may play a protective effect on the kidney tissuesby lowering the expression of IL-17which is one kind of pro-inflammatory cytokinesand reducing the extent of the inflammatory response and cell damage of themid-grade sepsis.
